矢车菊-3-葡萄糖苷对高脂饲料喂养小鼠血小板参数的影响 被引量:2

The in vivo effects of cyanidin-3-glucoside on platelet parameters in high fat fed mice

摘要 目的探讨桑葚提取物花色苷矢车菊-3-葡萄糖苷(Cy-3-g)对高脂饲料喂养小鼠血小板参数的影响。方法 8周龄健康C57BL/6J雄性小鼠,随机分为四组。AIN-93G组(Control)小鼠喂饲AIN-93G标准饲料、AIN-93G+Cy-3-g组(Control+Cy-3-g)喂饲添加桑葚提取物花色苷(1 000 mg/kg)的标准饲料;高脂组(HFD)喂饲高脂饲料;HFD+Cy-3-g组(HFD+Cy-3-g)喂饲添加桑葚提取物花色苷(1 000 mg/kg)的高脂饲料。实验周期为12周。实验周期结束后,检测小鼠血清血脂四项,使用血细胞分析仪进行血小板计数、平均血小板体积(MPV)、血小板分布宽度(PDW)测定;参照国际标准方法测定小鼠出血时间;噻唑橙标记网织血小板,使用流式细胞仪测定网织血小板所占外周血血小板百分比。结果与HFD组相比,HFD+Cy-3-g组小鼠的血清甘油三酯、总胆固醇、低密度脂蛋白含量降低,高密度脂蛋白含量升高,血小板数量升高,MPV及PDW降低(P<0.05)。与Control组相比,Cy-3-g可升高Control+Cy-3-g组小鼠的血清高密度脂蛋白含量,降低MPV及PDW(P<0.05)。结论在不影响出血时间的情况下,花色苷可以改善高脂饲料喂养小鼠的血小板参数异常变化。 Objective To estimate the effects of mulberry extract anthocyanin cyanidin-3-glucoside on platelet parameter in high fat feed mice. Methods The mice were divided into four groups randomly and fed with AIN-93 G, AIN-93G+Cy-3-g, HFD, HFD+Cy-3-g diets, respectively for 12 weeks. Mouse serum was collected for the detection of TC, TG,HDL, and LDL. Platelet count, mean platelet volume(MPV) and platelet distribution width(PDW) were measured by hematology analyzer. Bleeding time was tested through the cutting of mouse tail vein according to international normal method. The reticulated platelets were stained by thiazole orange and analyzed by flow cytometry to count the percentage of reticulated platelets. Results Compared with HFD group, the TC, TG, and LDL levels were lower in HFD +Cy-3-g group. The platelet count in HFD+Cy-3-g group was significantly higher than that of HFD group; the MPV and PDW in HFD+Cy-3-g group were lower than those of HFD group. Compared with AIN-93 G group,the HDL level, MPV, and PDW of AIN-93 G +Cy-3-g group were higher than those of AIN-93 G group. Conclusion Cy-3-g could ameliorate platelet parameters abnormal change induced by hyperlipidemia in vivo, without affecting bleeding time.
