木薯根系丛枝菌根真菌染色方法 被引量:4

A method to stain arbuscular mycorrhizal fungi in cassava roots

摘要 以大田木薯根为样品,探讨Quink牌纯黑墨水、北京牌蓝黑墨水、酸性品红、苏丹红Ⅳ、台酚蓝和苯胺蓝等6种染色剂对木薯根系丛枝菌根真菌染色效果的影响。结果表明,Quink牌纯黑墨水染色效果最好,用其染色的根系AM真菌的菌丝、泡囊、孢子等结构清晰可见。此方法操作简便且无毒,成本低廉,染色效果稳定,适用于木薯根系丛枝菌根真菌的染色和制片观察。 Taking the field cassava roots as a sample,Quink pure black ink,Beijing blue-black ink,acid fuchsin,Sudan red Ⅳ,trypan blue and aniline blue were applied to stain arbuscular mycorrhizal( AM) fungi of cassava roots and the dyeing effects were observed. The results showed that the dyeing effect of Quink pure black ink was the highest,and the AM fungal hyphae,vesicle,spore and other structures of roots which were stained with Quink pure black ink could be clearly seen. This method was simple,low toxicity,low cost and stable for staining AM fungi in cassava roots with excellent staining results.
