摘要
从奶粉中蛋白质的快速检测方法入手,选取了分光光度法中的考马斯亮蓝G-250来进行研究。结果表明,在酸性介质中,蛋白质与考马斯亮蓝G-250溶液反应生成蓝色络合物。该络合物最大吸收波长为594 nm,摩尔吸光系数为3.75×10^(-5)L/(mol·cm),精密度(RSD)为0.55%,相关系数为0.999 3,加标回收率为97.8%。
The rapid determination method of protein was studied. A Bradford G-250 method among spectrophotometric colorimetry was chosen to determine protein content in milk powder. The results showed that the maximum absorption wavelength of protein and Bradford G-250 red-brown generating blue complex was594 nm in the acidic medium and the apparent molar absorptive was 3.75 ×10^(-5) L/(mol·cm), the precision(RSD) was 0.55 %, the relevant coefficient was 0.999 3, the recovery of standard addition was 97.8 %.
