摘要
目的:建立HPLC同时测定润肠降脂胶囊中芦荟大黄素、大黄酸、大黄素、大黄酚及大黄素甲醚的含量测定方法。方法:采用Diamonsil C18(4.6mm×250mm,5μm)色谱柱;流动相:甲醇0.05%磷酸溶液,梯度洗脱(0-6min:80%-85%甲醇;6-22min:85%甲醇);检测波长:254nm;流速:1.0m L/min;柱温:30℃;进样量:10μL。结果:线性范围分别为:芦荟大黄素2.65-42.4μg/m L、大黄酸0.545-8.72μg/m L、大黄素2.55-40.8μg/m L、大黄酚12.60-201.6μg/m L和大黄素甲醚5.20-83.2μg/m L。平均加样回收率分别为:芦荟大黄素95.87%、大黄酸99.77%、大黄素100.15%、大黄酚97.52%、大黄素甲醚96.72%。结论:该法简单、快捷、重复性好,可作为润肠降脂胶囊的质量控制方法之一。
Objective: To establish an HPLC method for simultaneous determination of aloe-emodin, rhein, emodin, chrysophanol and physcion in Runchang Jiangzhi Capsule. Methods: Separation was performed on a Diamonsil C18(4.6mm×250mm, 5μm) column with a mobile phase consisting of methanol-0.05% phosphoric acid solution in gradient elution(0-6min: 80%-85% methanol, 6-22min: 85% methanol) at 30℃. The flow rate was 1.0m L/min, and the injection volume was 10μL. The detection wavelength was 254 nm. Results: The linear ranges of aloe-emodin, rhein, emodin, chrysophanol and physcion were 2.65-42.4, 0.545-8.72, 2.55-40.8, 12.60-201.60, and 5.20-83.2μg/m L, respectively. Their average recoveries were 95.87%, 99.77%, 100.15%, 97.52%, 96.72%, respectively. Conclusions: The method is simple, reliable, and reproducible. It could be used for the quality control of Runchang Jiangzhi Capsule.
