摘要
目的检测紫外光A/核黄素角膜交联(ultraviolet A/riboflavin corneal cross-linking,UVARCXL)后兔角膜基质内基质金属蛋白酶(matrix metalloproteinase,MMP)-1的短期含量变化。方法 15只新西兰白兔,随机分为3组,每组5只,A组为空白对照组,B组为角膜交联后3 d,C组为角膜交联后7 d。B、C两组行UVARCXL。各组取角膜后去除角膜上皮和内皮细胞,应用ELISA法检测角膜基质内MMP-1的含量。结果 A组角膜基质内MMP-1/总蛋白含量为(0.140±0.036)×10-6,B组为(0.242±0.059)×10-6,C组为(0.372±0.061)×10-6,3组之间差异有统计学意义(F=24.051,P=0.000)。UVARCXL后3 d,兔角膜基质内MMP-1含量显著升高,与A组相比差异有统计学意义(P<0.05),术后7 d其含量进一步升高,与B组相比差异有统计学意义(P<0.01)。结论 UVARCXL后7 d内,兔角膜基质内MMP-1含量持续增加,推测MMP-1可能对UVARCXL后的角膜基质重塑起一定作用。
Objective To determine the content changes of matrix metalloproteinase-1( MMP-1) in rabbit corneal stroma in the short term after ultrovialet A / riboflavin corneal cross-linking( UVARCXL). Methods Fifteen New Zealand white rabbits were random Ly divided into three groups,5 rabbits in each group. Group A: Control group; Group B: 3 days after UVARCXL; Group C: 7 days after UVARCXL. MMP-1 content in corneal stroma was measured by ELISA after removing the corneal epithelium and endothelium. R esults The proportion of MMP-1 relative to the total protein of group A was( 0. 140 ± 0. 036) × 10^- 6,group B was( 0. 242 ± 0. 059) × 10^- 6,and group C was( 0. 372 ± 0. 061) × 10^- 6. The difference among 3 groups was statistically significant( F = 24. 051,P = 0. 000). Three days after UVARCXL,MMP-1 content increased significantly,and the difference was statistically significant compared with group A( P 〈0. 05). Seven days after UVARCXL,the content of MMP-1 further increased,and the difference was statistically significant compared with group B( P 〈0. 01). Conclusion The content of MMP-1 in rabbit corneal stroma increase continuously in 7 days after UVARCXL,which means that MMP-1 may play a role in remolding corneal stroma after UVARCXL.
