摘要
目的正交试验优选复方清血片的最佳提取工艺。方法采用正交试验设计法,以落新妇苷、黄杞苷、总黄酮的含量和浸膏得率为指标,考察乙醇浓度、提取时间、加醇倍量3个因素对乙醇提取效果的影响;以总多糖、没食子酸的含量和浸膏得率为指标,考察煎煮次数、煎煮时间、加水倍量3个因素对水提取效果的影响。采用层次分析法确定综合权重,对实验结果进行综合评价。结果复方清血片醇提的最佳工艺为10倍量70%乙醇,提取2次,每次2 h;最佳水提工艺为:加10倍量水,提取3次,每次1 h。结论验证试验表明优选出的提取工艺稳定,合理,可行。
Objective To study extraction technology of compound Qingxue Tablets. Methods Orthogonal test was used to optimize the ethanol extraction technology by taking ethanol concentration, the amount of ethanol, extraction time and soaking time. With extraction content of astilbin, engeletin, total flavonoids and extract yield as comprehensive evaluation indexes. Orthogonal test was used to optimize the water extraction technology by taking the amount of water, extraction times and soaking time. With extraction content of polysaccharide, gallic acid and extract yield as comprehensive evaluation indexes. Analytic hierarchy process (AHP) was used to determine the weighted coefficient for optimization and synthetic evaluation of the multiple indexes. Results Orthogonal experimental design was applied to decide the optimal process as follows: extracting time 2 hours, extracting for 2 times, alcohol amount 10 times, alcohol concentration of 70%; Optimum water extraction technology conditions were as follows: extracted three times with ten times the amount of water, each time for 1 h. Conclusion The verification test showed that the optimum extraction process was stable, reasonable and feasible.
