摘要
目的:观察电针结合运动康复训练对兔膝关节软骨损伤模型关节液中IL-1β、TNF-α表达的影响。方法:按照随机数字表法将24只雄性新西兰大耳白兔随机分为空白对照组、模型组、运动康复组及电针结合运动康复组,每组6只。空白对照组不予任何处理,模型组进行兔膝关节软骨损伤模型制备,运动康复组在模型制备后给予电动跑台训练,电针结合运动康复组在运动康复组基础上增加电针治疗。治疗4周后,各组兔膝关节软骨进行大体观察及HE染色,并采用Elisa法检测关节液中IL-1β、TNF-α的含量。结果:治疗4周后,与空白对照组比较,模型组兔膝关节软骨损伤明显,存在明显炎性细胞浸润现象,关节液中IL-1β、TNF-α的含量明显升高(P<0.05);与模型组比较,运动康复组和电针结合运动康复组兔膝关节软骨损伤得以改善,炎性细胞浸润现象有所缓解,关节液中IL-1β、TNF-α的含量均明显减少(P<0.05);与运动康复组比较,电针结合运动康复组兔膝关节软骨损伤程度进一步降低,膝关节液中IL-1β、TNF-α的含量有所下降(P<0.05)。结论:4周的电针结合运动康复训练可明显降低兔膝关节软骨损伤模型关节液中IL-1β、TNF-α的含量,改善关节软骨的病理性损伤,疗效优于单纯运动康复训练。
Objective: To observe the effect of electro- acupuncture combined with exercise rehabilitation training on the expression of IL- 1β,TNF- α in joint fluid of rabbits with the injury of knee articular cartilage.Methods: 24 male New Zealand white rabbits were randomly divided into a Control group( C),a Model group( M),an Exercise Rehabilitation Training group( ERT) and an Electro- acupuncture( EA) combined with ERT group( EA + ERT). Control group was without any intervention. The rabbits in Model group,ERT group and EA + ERT group were made into the models with injury of knee cartilage. ERT group was treated by exercise rehabilitation training. EA + ERT group was treated by EA combined with ERT. Then the observation was conducted by using HE staining,and the content of IL- 1β,TNF- α in knee joint fluid of these four groups was detected by using Elisa after the treatment was given for 4 weeks. Results: The contents of IL- 1β,TNF- α in knee joint fluid of ERT group and EA + ERT group were both significantly reduced. Compared with the Model group,the difference was statistically significant( P 〈0. 05). Meanwhile,the damage of knee articular cartilage was improved. Compared with ERT group,the contents of IL- 1β,TNF- α in knee joint fluid of EA + ERT group were reduced( P〈 0. 05). Conclusion: Electro- acupuncture combined with exercise rehabilitation training can reduce the contents of IL- 1β,TNF- α in knee joint fluid and can improve pathological articular cartilage damage,which is more effective than exercise rehabilitation training alone.
