摘要
以裸燕麦为原料,采用Osborne蛋白分级方法分离提取裸燕麦谷蛋白,利用蛋白酶水解后以水解度和对·OH清除率为指标,通过正交试验对酶解裸燕麦谷蛋白的工艺进行探索。SDS-PAGE电泳结果显示裸燕麦谷蛋白分子量范围在(20~97)ku,且最佳水解蛋白酶为碱性蛋白酶。谷蛋白酶解最佳工艺为:底物浓度10 g/L,酶用量10%,p H值8.5,温度55℃。在最佳酶解条件下水解度近40%,·OH清除率达60%以上。
Took naked oats as raw material, separated glutelin through Osborne, then hydrolyzed by protease. DH and scavenging -OH as index, orthogonal test were taken to hydrolyze naked oats glutelin. SDS-PAGE electrophoresis showed that naked oats glutelin molecular weight ranged from 20 ku to 97 ku, alkaline was the better one to hydrolyze glutelin. The obtained optimum glutelin hydrolysis parameters were as follows: concentration of substrate 10 g/L, dose of enzyme 10%, pH8.5, temperature 55 %. With these parameters, the DH reached nearly 40%, the ratio of scavenging .OH could reach higher than 60%.
