摘要
探讨桔梗抗肺炎支原体(MP)有效部位对感染大鼠肺组织中TGF-β1 m RNA表达的影响.采用滴鼻法构建肺炎支原体肺炎大鼠模型,用光学显微镜观察大鼠肺组织形态学的变化,运用RTPCR法检测TGF-β1在空白对照组、模型对照组、阿奇霉素组、桔梗有效部位高、中、低剂量组的表达情况.结果表明,空白组大鼠的肺组织结构正常,模型组大鼠的肺组织呈重度或中度炎症病变,造模成功,给药各组大鼠的肺组织病变均有所改善.模型组大鼠肺组织中TGF-β1 m RNA的表达量明显增多,桔梗抗肺炎支原体有效部位高、中、低剂量组和阳性药组均能下调感染MP大鼠肺组织中TGF-β1的表达,其中阿奇霉素组和桔梗有效部位高剂量组效果最佳.桔梗有效部位对MP感染致大鼠肺损伤的修复作用机制可能是通过下调TGF-β1,抑制了上皮间质转化的过程.
To investigate the effects of active parts isolated from Platycodon grandiflorum on the expression of TGF- β1 m RNA in rats infected with mycoplasma pneumonia. The model of rats infected with mycoplasma pneumoniae was established by nasal drip to observe the pathological changes extent of rats' lung tissue by light microscope. The levels of TGF- β1m RNA in pulmonary tissues were determined with RT- PCR. Results showed that the control group rats' lung tissue structure were normal; model group rats' showed severe or moderate inflammatory lesions,which predicted building model success; drug groups can reduce the lung inflammation and the lung tissue lesions were improved. TGF- β1 m RNA of the model group significantly increased than the coutrol group; medicated groups can reduce the expression of TGF- β1 m RNA,while the effect of positive medicine group and the high dose group were better than others. The anti MP mechanism of Platycodon grandiflorum active parts was associated with downregulation of the expression of TGF- β1 m RNA and inhibiting the process of epithelial- mesenchymal transition.
