摘要
目的:观察加味青娥丸对糖尿病SD大鼠糖脂代谢和其肾功能的影响,并对其可能机制进行初步探讨。方法:选取月龄一致的正常健康SD大鼠,将其随机分为加味青娥丸低剂量组、加味青娥丸中剂量组、加味青娥丸高剂量组,并设置正常对照组、模型组。通过对其喂养高脂饲料和腹腔注射25 mg/kg的链脲菌素(STZ)来诱导建立SD大鼠的2型糖尿病模型。每组分别给予相应的药物、生理盐水进行灌胃,连续8周。测定各组大鼠的空腹血糖(FBG)、餐后2 h的血糖(2PBG)、肾脏质量指数(Kw/Bw)以及24 h的尿蛋白含量,并于最后一次给药后2 h将大鼠处死,并应用生化仪对血清中甘油三酯(triglyceride,TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(low densitylipoprotein cholesterol,LDL)、高密度脂蛋白胆固醇(high densitylipoprotein cholesterol,HDL)、血肌酐(SCr)、尿素氮(BUN)进行测定。应用ELISA法对血清中IL-1β、MCP-1、超敏c反应蛋白(hs-CRP)及CCL5的含量进行测定。结果:观察发现,与模型组大鼠相比,给药组大鼠的FBG、2PBG、24 h尿蛋白的含量、Kw/Bw,血清中TG、TC、LDL、SCr、BUN含量都明显降低,HDL含量增高。同时,给药组也能降低血清中IL-1β、MCP-1、hs-CRP及CCL5的含量。结论:加味青娥丸能够降低2型糖尿病SD大鼠的血糖、血脂,并且对其肾功能也有一定的改善,此作用可能与降低血清中IL-1β、MCP-1、hs-CRP及CCL5的表达有关。
AIM: To observe the effectiveness of modified Qing'e pill on glucolipid metabolism and renal function in type 2 diabetic rats. METHODS: Rats of same months were randomly divided into five groups: modified Qing'e pill low dose group, modified Qing'e pill middle dose group, modified Qing'e pill high dose pills, control group and model group. High-fat diet combined with intraperitoneal injection of 30 mg/kg STZ were given to make model of type 2 diabetes. Each group was administered with respective drug and saline lavage for 8 weeks. FBG, 2PBG, Kw/Bw and 24 hours urinary protein content in each group were measured, Biochemical analyzer was applied to measure TG, TC, LDL, HDL, SCr, BUN. ELISA method was employed to measure the contents of IL-1β, MCP-1, hs-CRP, CCL5 in serums. RESULTS: Compared with model group, FBG, 2PBG, Kw/Bw, 24 hours urinary protein content, TG, TC, LDL, SCr, BUN,IL-1β,MCP-1 ,hs- CRP, CCL5 all reduced while HDL levels were detected increased in administered groups. CONCLUSION: Modified Qing'e pill can reduce blood sugar, Lipid, and also improve renal function in type 2 diabetes rats. Its protective mechanisms may in part associated with the decreasing of IL-1β, MCP-1, hs-CRP ,CCL5.
