鱼腥草总黄酮与利血生抗辐射功效的对比研究 被引量:9

Comparative Study on Total Flavonoids of Houttuynia Cordata and Leucogen Anti Radiation Effect

摘要 目的比较鱼腥草提取物鱼腥草总黄酮与利血生对X射线辐射损伤大鼠骨髓造血功能和免疫功能的影响。方法建立模型:给予大鼠剂量为4 Gy、剂量率为2 Gy/min的X射线照射,成功制备了X射线损伤模型。将造模成功的大鼠随机分为4个组:分别为对照组(模型组)、全成分提取液组(全成分,A组)、鱼腥草总黄酮水提醇溶部位(含总黄酮20.15%,B组)、利血生组(12.6 mg/kg的利血生混悬液,C组),每组各8只。分别给予等体积的蒸馏水、鱼腥草的全成分提取液(100 g鲜药/kg)、总黄酮水提醇溶部位(5.23 g/kg)、12.6 mg/kg的利血生混悬液,正常对照组大鼠8只,给予等体积的蒸馏水灌胃。给药期间,每周对大鼠眼眶取血检测血常规,给药63天后,处死大鼠,检测免疫系统、骨髓造血系统以及抗氧化的相关指标。结果给予鱼腥草不同成分提取物与利血生灌胃63天后,观察到:1A组鱼腥草全成分提取液可提高大鼠的胸腺和脾脏指数;增加大鼠外周血网织红细胞数;降低外周血网织红细胞微核率;降低骨髓嗜多染红细胞微核率;提高CAT和GSH-Px含量。2B鱼腥草总黄酮组能显著提高辐射损伤大鼠的免疫力,增强辐射损伤大鼠的骨髓造血功能,并能增加SOD及GSH-Px的含量,增强机体的抗氧化作用;增加白系细胞数和网织红细胞数,升高脾脏指数,增加骨髓DNA含量,降低外周血和骨髓微核率。3C组利血生可提高大鼠的胸腺和脾脏指数;对射线损伤大鼠白细胞数和中性粒细胞数的影响与鱼腥草总黄酮相近;不能显著升高辐射损伤大鼠的骨髓DNA含量、SOD含量及GSHPx含量,抗辐射氧化损伤的作用较弱。结论鲜鱼腥草全成分提取液在一定程度上可提高辐射损伤大鼠的免疫力,保护辐射损伤大鼠的骨髓造血系统;鱼腥草总黄酮可显著提高辐射损伤大鼠的免疫力,增强辐射损伤大鼠的骨髓造血系统功能;利血生有一定的提高X射线损伤大鼠白细胞数和中性粒细胞数作用,但综合比较利血生的抗辐射作用明显弱于鱼腥草总黄酮。鱼腥草可以部分替代利血生的作用。 Objective Compared of fresh Herba Houttuyniae extract( traditional Chinese Medicine) of total flavonoids from Houttuynia cordata and leucogen( Western Medicine) of X- ray radiation damage effects of rat bone marrow function and immune function. Methods 62 male SD rats,of which 15 were sham radiation,as normal( blank)control group; 47 for 4Gy X- ray irradiation. The model was composed of 15 normal control group and 5 X- ray radiation model. The number of white blood cells,neutrophils and lymphocytes were detected. The results showed that the model was successful. The rest of the successful model of 32 rats were randomly divided into 4 groups: model control group( model group),full component extraction solution group( Total composition packet),total flavone group,leucogen group,each group 8 rats in each. SLN were poured into the stomach volume of distilled water,Houttuynia cordata composition extraction liquid( 100 g fresh drug / kg),total flavonoids extract 5. 23 g / kg and 12. 6mg / kg the Leucogen suspension gavage. In normal conrmal group,10rats( 8 rats) were given the same volume of distilled water. During administration,a week to the eyes of rats from routine blood testing observed flavonoids from Houttuynia cordata Thunb and benefit blood born and number of red blood cells of rat leukocyte,neutrophil cells,lymphocytes,network fabric. After 63 days,the rats were killed,and the immune system,bone marrow hematopoietic system and the related indexes were detected. Results The dose of X was 4 Gy and the dose rate was 2 Gy / min,and the X ray damage model was successfully prepared.Given Houttuynia components of different extract by intragastric 63 days observed the 1Houttuynia cordata composition extracted liquid in a certain extent can improve the immunity of rhe rat rediation damage and radiation protection injury of rat bone marrow hematopoietic system. 2The total flavonoids of Houttuynia cordata can significantly improve the radiation damage of the immunity of rats,To enhance the anti- oxidation function of organisms. Can increase the content of SOD and GSH- Px,enhance the body's antioxidant effect. 3leucogen can significantly increase the thymus index and spleen index of rats; to radiation injury rat white blood cell count and neutrophil cell count and the influence of the flavonoids from Houttuynia cordata thumb similar; failed to significantly elevate the radiation injury of rat bone marrow DNA content,the content of SOD and GSH PX content,anti radiation oxidative damage effect is weak. Conclusions Fresh Houttuynia leaf has good radioresistance effect,can be widely used,improve economic effciency. The total flavonoids of Houttuynia cordata can significantly improve the radiation injury of rat immunity,enhance the radiation function of bone marrow hematopoietic system injury in rats. leucogen with enhanced X- ray injury rat white blood cell count and neutrophil cell count role,but compared with Leucogen Antiradiation Effect of synthesis significantly weaker in the flavonoids from Houttuynia cordata thumb.
