药物跨血脑屏障转运的实验模型研究进展 被引量:4

摘要 目的介绍血脑屏障(blood-brain barrier,BBB)的细胞结构及神经系统药物BBB渗透性评价的体内、外模型,为药物跨BBB转运研究开发提供理论基础和实验技术参考。方法综述近10年中外文献中有关BBB定义、细胞组成、结构特点及药物渗透BBB的体内、外评价模型研究状况。结果介绍了BBB的细胞组成和结构特点,评价了体外非细胞和细胞模型及体内BBB渗透性模型的评价方法,介绍了各个模型的优缺点及适用环境。结论目前尚无化合物BBB通透性体内、外评价的"金标准"模型,应建立、使用合理优化的细胞模型,结合动物体内BBB渗透实验结果,描绘出药物的BBB渗透特点的轮廓。 Objective To introduce the structure of blood-brain barrier(BBB)and drug evaluation models,and provide basic theory and experimental techniques for the evaluation of novel central nervous system(CNS)drugs. Methods The literature in recent 10 years on the BBB definition,structural characteristics and drug screening BBB models in vivo and in vitro were reviewed. Results The composition and structural characteristics of the BBB were described,and the advantages and disadvantages of each one of these models were evaluated. Conclusion So far,there is no "gold standard" model for estimating the permeability of a compound on BBB,the optimized cell models should be combined with the BBB permeability models in vivo to predict the extent of BBB permeability of a compound.
