摘要
从正常笼养树鼩腹部的脂肪转录组数据开发SNP分子标记。检测了6只动物共计274 278 568条unigene(总长度43 138 417 bp)序列信息后,在262 980条unigene(5.75%)中发现SNP位点263 071个,SNP发生频率为1/164 bp,其中转换(transition)177 562个,颠换(transversion)85 509个。在所有变异类型中,A/G和C/T发生频率最高,分别达33.85%和33.66%。将包含SNP位点的262 980条unigene通过参比序列进行注释,并对其进行GO分类、COG分类注释和代谢通路注释(KEGGpathway),结合已有研究,分别筛选到1 187个有GO注释、77个有COG注释和1 080条个有KEGG通路注释的脂肪转录组中的可能与脂肪形成和代谢有关的SNP标记。
SNP markers were developed from the transcriptome of tree shrew( Tupaiabelangeri) abdominal adipose tissues among 6 different normal healthy tree shrews. After detecting 274 278 568 unigenes( 43 138 415 bp),a total of 263 071 SNPs were identified in 262 980 unigenes( 5. 75%) with the frequency of 1 /164 bp,including transition177 562 and transversion 85 509. A / G and C / T are the main types,accounting for 33. 85% and 33. 66% of all the SNPs,respectively. All the 262 980 SNP of all the unigenes were functionally annotated using the Gen Bank Nrdatabas,geneontology( GO),clusters of orthologous groups( COG) and KEGG pathways. There are 1 178 SNPs annotated by GO,77 SNPs annotated by COG and 1 080 annotated by KEGG,which may link with the key genes of adipose formation and metabolism in adipose tissue,were obtained.
