摘要
目的采用星点设计-效应面法优化苦豆子生物碱减压提取工艺。方法采用单因素和星点设计-效应面法相结合,以苦豆子总碱及氧化苦参碱的含量为考察指标,优选苦豆子生物碱的提取工艺。结果确定最佳提取工艺为温度60℃,溶剂用量10倍,提取时间90 min,提取3次;提取工艺验证结果与二次项拟合方程预测值的偏差为-2.144%,偏差较小(<5%)。结论优选的减压动态提取工艺稳定可行,预测性良好。
Objective To optimize the vacuum extraction of alkaloids from Sophora alopecuroides by central composite design-response surface methodology. Methods Combine with single factor test and central composite design-response surface methodology ,the vacuum extraction technology was optimized,the contents of total alkali and oxymatrine were determined. Results The condition for optimum extraction process was 60 ℃,adding 10-fold solvent and extract for 90 min extration three times.Deviation between extraction validation result and predicting value from second-order equations was -2.144%,which was a small deviation (〈5%). Conclusion The optimized vacuum extraction method of alkaloids from Sophora alopecuroids is stable,feasible and well predictive .
