摘要
目的探讨针加灸防治阿尔茨海默病(AD)的作用机制。方法 60只Wistar大鼠随机分为4组:正常组、模型组、假手术组及针加灸治疗组。大鼠海马注射Aβ1-42复制AD模型,在百会和肾俞(双)进行针刺加灸,用ELASA法检测血清Aβ水平,用免疫组化方法检测大鼠海马CA1区APP蛋白的表达。结果模型组血清Aβ浓度,海马APP的表达比正常组、假手术组显著升高(P<0.05),针加灸治疗组与模型组比较,能显著降低血清Aβ及海马APP的表达,有显著差异(P<0.01)。结论 "益肾调督"针加灸法可能通过减少或抑制AD大鼠血清内Aβ水平,降低大鼠海马CA1区APP的表达,延缓阿尔茨海默病的病理进程,对AD起着有效的防治作用。
Objective To study the impact of Supplementing Kidney and Strengthening Du Meridian Acupuncture Method on AD rats' A beta level and its precursor Protein APP,and explore the mechanism of prevention and treatment of AD by reducing the Aβlevel and APP. Methods 60 Wistar rats are randomly divided into 4 groups: normal group,model group and sham group and the treatment group,A beta 1-42 was injected in rats' hippocampal which was used to replicate model. Acupuncture and moxibustion on AD rats' Baihui and Shenshu was used to treat the AD rats,ELISA was used to detect serum Aβ levels,APP levels in rat hippocampal CA1 region by immunohistochemical method to detect. Results( 1) AD rats 'serum Aβ concentrations between groups: the difference between treatment group and model group was statistically significant( P〈 0. 01),sham group and model group difference was statistically significant( P 〈0. 05).( 2) AD rat hippocampal levels of APP: there is no significant difference between sham group and the normal group( P〈0. 05),model group and the normal group have a more significant difference( P〈 0. 05),difference between treatment groups in the model group was significantly( P〈 0. 01). Conclusion Aβ1-42 injection prepared AD model successfully,Supplementing Kidney and Strengthening Du Meridian Acupuncture Method can reduce or inhibit the AD rat serum'A beta level and reduce AD rats' hippocampus CA1 area APP,delay the pathological process of alzheimer's disease,treat and prevent AD effectively.
