摘要
为了明确人工栽培桑黄(Phellinus spp.)的药用价值,以野生桑黄和人工栽培桑黄的子实体为材料,检测分析其水溶性活性物质粗多糖和粗酚的含量,以及其抗肿瘤、抗感染活性。采用乙醇浸提及甲醇萃取的方法提取栽培桑黄子实体粗酚的得率显著高于野生桑黄子实体,经超高效液相色谱-飞行时间质谱(UPLC-MS)技术分析2种桑黄子实体中的桑黄酚主要由丁香酸、原儿茶酸、原儿茶醛、咖啡酸4种单体酚组成。采用MTT法检测2种桑黄子实体来源粗多糖、粗酚以及4种单体酚物质对体外培养HCT116、HT-29、HepG 2肿瘤细胞的增殖均有较强抑制作用,2种来源的活性成分样品在相同剂量下的抑制作用差异不显著,其中原儿茶醛抑制肿瘤细胞增殖的活性较强。用流式细胞术体外检测桑黄粗酚中的单体酚物质对结肠炎患者外周血CD4+T细胞凋亡具有诱导活性,且咖啡酸、原儿茶酸抑制细胞增殖并诱导其凋亡的活性较强。研究结果表明,人工栽培桑黄子实体有良好的药用价值,其中多糖和原儿茶醛成分是抑制结肠癌、肝癌细胞增殖的主要活性物质,丁香酸、原儿茶酸通过诱导CD4+T细胞凋亡发挥抗感染作用。
In order to explore the medicinal value of cultivated Phellinus spp.,wild and cultivated Phellinus spp. fruit bodies were used as materials to determine contents of water soluble substances including crude polysaccharides and crude polyphenols,and to investigate their anti-tumor and anti-inflammatory activities. The results showed that the extraction rates of crude polyphenols from cultivated Phellinus spp. fruit bodies were significantly higher than those from wild Phellinus spp. fruit bodies. UPLC-MS analysis revealed that the crude polyphenols from wild and cultivated Phellinus spp. fruit bodies were mainly composed of syringate,protocatechuate,protocatechualdehyde and caffeic acid. MTT as-say demonstrated that crude polysaccharides,crude polyphenols and individual polyphenols( syringate,protocatechuate,protocatechualdehyde,and caffeic acid) extracted from both wild and cultivated Phellinus spp. fruit bodies had strong inhibitive effects on proliferation of HCT116,HT-29,and Hep G2 cancer cells,and there was no significant difference between them at the same dose.Moreover,protocatechualdehyde had stronger anti-tumor activity than other ingredients. After the CD4 + T cellsfrom patients with inflammatory bowel diseases( IBDs) were treated with different polyphenols,cell apoptosis was observed by flow cytometry. Moreover,caffeic acid and protocatechuate had stronger activity in inhibiting cell proliferation and inducing cell apoptosis. These results indicate that the cultivated Phellinus spp. fruit bodies have promising medicinal value,in which the polysaccharides and protocatechualdehyde have strong anti-colon cancer and anti-liver cancer activity,and syringrate and protocatechuate play anti-inflammatory role by inducing apoptosis of CD4 + T cells.
