芹菜素纳米乳抗大鼠肝纤维化作用及对MMP-1,TIMP-1的影响 被引量:3

Effects of apigeninnanoemulsion on hepatic fibrosis and tissue levels of MMP-1,TIMP-1 in rats

摘要 目的:观察芹菜素纳米乳对CCl4诱导大鼠肝纤维化模型的作用及对基质金属蛋白酶-1(MMP-1)、金属蛋白酶抑制因子-1(TIMP-1)的影响。方法:采用CCl4复制大鼠肝纤维化模型,自造模之日开始,灌胃给药,1次/d,连续8周。末次给药后24 h,取大鼠血和肝脏,观察芹菜素纳米乳对血清肝功能指标(ALT、AST)、肝纤维化指标(HA、LN、Ⅲ-C、Ⅳ-C)和肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、羟脯氨酸(Hyp)、MMP-1、TIMP-1水平的影响。结果:与模型组比较,各给药组肝功能和肝纤维化均有不同程度的改善。结论:芹菜素纳米乳对CCl4诱导肝纤维化大鼠具有一定的治疗作用,其作用可能与抗脂质过氧化反应、影响MMP-1和TIMP-1表达有关。 AIM: To observe therapeutic effects of apigeninnanoemulsion on CCl4 induced hepatic fibrosis and hepatic tissue level of MMP-1,TIMP-1 in rats. METHODS: The model of rat liver fibrosis was adopted which was induced by CCl4 injection.Drugs were administrated from the first day for 8weeks,once a day. 24 hours after the last administration,the levels of hepatic function indices( ALT,AST) and hepatic fibrosis indices( HA,LN,Ⅲ-C,Ⅳ-C) in serum and MDA,Hyp,SOD,GSH-Px,MMP-1 and TIMP-1 activity in hepatic tissue were detected. RESULTS: Compared with the model group,liver function and hepatic fibrosis were improved in drug groups. CONCLUSION: Apigeninnanoemulsion has a significant effect on the liver fibrosis rats,its mechanism is probably related to resisting-lipid peroxidation and adjusting the expression of MMP-1 and TIMP-1.
