知柏地黄汤对解脲脲原体感染模型大鼠精子线粒体NADH脱氢酶mRNA、ATP合成酶mRNA表达的影响

摘要 目的研究知柏地黄汤对解脲脲原体(UU)感染模型大鼠精子线粒体NADH脱氢酶mRNA、ATP合成酶mRNA表达的影响。方法取体质量为(220±10)g雄性SD大鼠80只,随机分成假手术组、模型组、中药组(知柏地黄汤)、西药组(强力霉素)、中西药组(知柏地黄汤+强力霉素),造模成功后,假手术组与模型组给予等量的生理盐水灌胃,治疗组(中药组、西药组和中西药组)给予相应的药物灌胃,连续用药21d。末次给药24h后取标本,采用常规精液检测法检测精液质量,RT-PCR法检测精子NADH脱氢酶mRNA、ATP合成酶mRNA表达。结果假手术组、模型组、中药组、西药组、中西药组大鼠精子NADH脱氢酶mRNA表达水平分别为0.44750±0.017078、0.24750±0.015000、0.34250±0.017078、0.36500±0.012910、0.39400±0.012100;ATP合成酶mRNA表达水平分别为0.36500±0.012910、0.27250±0.009574、0.35250±0.018930、0.34750±0.017078、0.39500±0.012910;直线速度分别为(12.93±1.08)μm/s、(6.24±1.13)μm/s、(12.78±0.92)μm/s、(10.43±1.32)μm/s、(13.54±1.16)μm/s;平均速度分别为(20.32±2.08)μm/s、(12.48±3.54)μm/s、(20.48±2.16)μm/s、(17.12±3.15)μm/s、(21.72±1.83)μm/s。中西药组NADH脱氢酶mRNA、ATP合成酶mRNA表达水平与模型组、中药组、西药组比较均有上升,差异均有统计学意义(P<0.01),中西药组精子运动速度与模型组、中药组、西药组比较均上升,差异均有统计学意义(P<0.01)。结论 UU感染大鼠模型精子运动速度及NADH脱氢酶mRNA、ATP合成酶mRNA表达水平变化,提示其可能是UU感染所致不育症的机制之一。 Objective To investigate the effects of Zhibai Dihuang Decoction on the expressions of sperm mitochondrial NADH dehydrogenase and ATP synthase in rat model of Ureaplasma urealyticum (Uu) infection. Methods Total of 80 male SD rats with body mass of (220+10)g were randomly divided into sham operation group, model group, Chinese medicine group (Zhibai Dihuang Decoction), western medicine group (doxycycline), western group (Zhibai Dihuang Decoction plus doxycycline), After rat models were successfully established, rats in the sham operation group and the model group were given equal amount of normal saline, and rats in the treatment group were intragastrically given the corresponding medicine, and the drug was administered continuously for 21 days. During 24 hours after the last administration, semen samples were collcetd, and the semen quality was detected by routine semen detection. RT-PCR was used to detect the expression of NADH dehydrogenase and ATP synthase in sperm. Results The expression levels of sperm NADH dehydrogenase in the control group, model group, Chinese medicine, western medicine group, the group of Chinese and western of rat were 0.44750±0.017078, 0.24750±0.015000, 0.34250±0.017078, 0.36500±0.012910, and 0.39400±0.012100 respectively; The expression levels of ATP synthetase were 0.36500±0.012910, 0.27250±0.009574, 0.35250±0.018930, 0.34750±0.017078 and 0.39500±0.012910 respectively; Linear velocity were (12.93±1.08)μm/s, (6.24±1.13)μm/s, (12.78±0.92)μm/s, (10.43±1.32)μm/s, (13.54±1.16)μm/s; Average speed of (20.32±2.08)μm/s, (12.48±3.54)μm/s, (20.48±2.16)μm/s, (17.12±3.15)μm/s, (21.72±1.83)μm/s. the expression levels of NADH dehydrogenase and ATP synthase in Chinese and western group level were significant increased compared with that in the model group, Chinese medicine group, western medicine group (P&lt;0.01), and sperm motility, vitality, motion velocity were also remarkably increased, the differences were statistically significant (P〈0.01).Conclusion The change of sperm motility rate and the expression of NADH dehydrogenase and ATP synthase may be one of the mechanisms of UU infection-caused infertility.
