乳腺癌中HER-2表达与其基因启动子甲基化的关系

摘要 目的:探讨女性乳腺癌组织中人表皮生长因子受体-2(HER-2)表达与其基因启动子甲基化的关系。方法:应用实时荧光定量PCR方法检测38例乳腺癌组织中HER-2启动子甲基化情况,并结合临床病理资料进行分析。结果:HER-2阴性组的HER-2基因启动子甲基化水平显著高于阳性组(P<0.05)。乳腺癌患者年龄、淋巴结转移、组织学分级及ER与HER-2基因甲基化无明显相关性(P>0.05);乳腺肿瘤大小与HER-2基因甲基化水平呈负相关(r=-0.6036,P<0.05)。结论:HER-2基因甲基化抑制了乳腺癌组织HER-2表达。 Objective:To investigate the relationship between the expression of human epidermal growth factor receptor-2(HER-2)and the promoter methylation of HER-2in female breast cancer.Methods:Using real-time fluorescence quantitative PCR method to detect the methylation status of HER-2promoter in 38 cases of breast cancer tissues,and to analyze the clinical pathological data.Results:The methylation level of HER-2gene promoter in HER-2negative group was significantly higher than that in the positive group(P〈0.05).There was no significant correlation between age,lymph node metastasis,pathological grade,ER and HER-2gene methylation in breast cancer patients(P〉0.05).Breast tumor size was negatively correlated with methylation level of HER-2gene(r=-0.604,P〈0.05).Conclusion:Methylation of HER-2gene inhibits the expression of HER-2in breast cancer tissues.
