UPLC法分析藤黄炮制前后4种成分的变化 被引量:3

摘要 目的建立UPLC法分析藤黄Garcinia hanburyi Hook.f.及其炮制品(高压制、豆腐制、清水制)中藤黄烯酸、表藤黄烯酸、藤黄酸、新藤黄酸含有量的变化。方法该植物乙腈提取液的分析采用UPLC HSS T3色谱柱(100 mm×2.1 mm,1.8μm);流动相乙腈-0.1%甲酸(80∶20);体积流量0.3 m L/min;检测波长360 nm。结果 4种成分在各自范围内均呈良好的线性关系(r〉0.999 3),平均加样回收率100.9%~103.0%(RSD〈3.0%)。炮制后,藤黄酸和新藤黄酸的含有量均有所下降,分别在清水、豆腐制品中最低。同时,新生成了藤黄烯酸和表藤黄烯酸,两者含有量在清水制品中最高,高压制品中最低。结论清水制法有利于藤黄烯酸和表藤黄烯酸的生成。 AIM To establish a UPLC method for analyzing the variations of contents of gambogellic acid,epigambogellic acid,gambogic acid and neogambogic acid in Garcinia hanburyi Hook. f. and its processed products( high pressure-processed,tofu-processed and water-processed). METHODS The analysis of acetonitrile extract of this plant was performed on a UPLC HSS T3 column( 100 mm × 2. 1 mm,1. 8 μm),with the mobilephase comprising of acetonitrile-0. 1% formic acid at a flow rate of 0. 3 mL / min,and the detection wavelength was set at 360 nm. RESULTS The four constituents showed good linear relationships within their own ranges( r 〉0. 999 3),whose average recoveries were 100. 9%-103. 0%( RSD 〈 3. 0%). After processing,the contents of gambogic acid and neogambogic acid were decreased,which were the lowest in water-processed and tofu-processed products,respectively. Simultaneously,gambogellic acid and epigambogellic acid were newly generated,whose contents were the highest in water-processed product,and the lowest in high pressure-processed product. CONCLUSION Processing with water is beneficial to the generations of gambogellic acid and neogambogic acid.
