摘要
2型糖尿病主要表现为胰岛素抵抗,即机体对胰岛素的敏感性降低。近年来,建立体外胰岛素抵抗细胞模型,来模拟机体胰岛素抵抗已成为研究热点。肝脏、脂肪组织和骨骼肌是胰岛素作用的主要靶器官。采用高胰岛素浓度、高糖高脂和地塞米松等体外诱导Hep G2细胞、3T3-L1细胞和C2C12细胞,观察细胞生长情况及葡萄糖摄取率的变化,建立胰岛素抵抗细胞模型,从而筛选具有改善胰岛素抵抗的中西药物。本文主要对糖尿病发病现状、胰岛素抵抗细胞模型建立方法、潜在改善胰岛素抵抗的药物评价做简要概述。
Insulin resistance is the characteristic of type Ⅱ diabetes, which means the organism shows a low level response to physiological effect of insulin. In recent years, cell model which represents insulin resistance in vitro to simulate the pathogenesis of insulin resistance inside organisms has become more popular. The main target cells that insulin works on are liver, adipose tissue, and skeletal muscle tissue. Methods such as high insulin concentrations, high sugar, high fat, and dexamethasone can induce HepG2 cells, 3T3-L1 cells, and skeletal muscle cells in vitro. The growth of cells and the changes of glucose uptake ratio are observed in establishing an insulin resistance cell model. Consequently, traditional Chinese and western drugs are able to be screened to improve the insulin resistance. This paper is mainly a brief overview of the present situation of diabetes, the methods of establishinginsulin resistance model, and evaluation of potential drugs inimproving insulin resistance.
