摘要
目的:探讨PIK3CA基因表达与子宫内膜异位症相关性卵巢癌的表达及意义。方法:用实时定量PCR方法检测100例子宫内膜异位症及60例子宫内膜异位症相关性卵巢癌中PIK3CA的m RNA水平,并选取正常卵巢组织20例作为对照;蛋白质免疫印迹(Western Blot)方法检测子宫内膜异位症及子宫内膜异位症相关性卵巢癌PIK3CA的蛋白水平;测序检测PIK3CA基因的热点变异区是否存在变异。结果:PIK3CA在子宫内膜异位症相关性卵巢癌中的m RNA水平明显高于子宫内膜异位症及正常卵巢组织(P<0.01);PIK3CA的蛋白表达产物在子宫内膜异位症相关性卵巢癌中呈强阳性表达,与子宫内膜异位症比较,差异具有统计学意义(P<0.05);PIK3CA在子宫内膜异位症相关性卵巢癌组织I期/II期之间、II期与III至IV期之间比较,差异具有统计学意义(P<0.05);PIK3CA的阳性表达与分化程度(P<0.01)、淋巴结转移(P<0.05);PIK3CA的3个热点突变区H1047R、E542K、E545K在子宫内膜异位症及子宫内膜异位症相关性卵巢癌中的突变频率差异无统计学意义(P>0.05)。结论:子宫内膜异位症相关性卵巢癌的发生与PIK3CA基因突变不相关;但PIK3CA基因表达水平与子宫内膜异位症相关性卵巢癌的恶性演变过程具有相关性。
Objective We aimed to investigate the association between PIK3 CA gene expression and endometriosisassociated ovarian carcinoma. Methods PIK3 CA m RNA expression levels of tissue blocks were measured by Real TimePCR, which were from 100 patients diagnosed with endometriosis, 60 patients diagnosed with endometriosis-associated ovarian carcinoma, and 20 healthy volunteers. Western Blot was employed to detect the PIK3 CA protein expression of endometriosis patients or endometriosis-associated ovarian carcinoma patients. Sequencing analysis was done to investigate the ‘hotspot＇mutation of PIK3 CA. Results The m RNA level of PIK3 CA from endometriosis-associated ovarian carcinoma was significantly higher than both endometriosis and healthy tissue（P〈 0.01）. Endometriosis-associated ovarian carcinoma samples exhibited positive expression of PIK3 CA, compared with endometriosis samples（P〈 0.05）. The expression of PIK3 CA between I and II, II and III-IV clinical stage of endometriosis-associated ovarian carcinoma had statistical significance（P 〈0.05）. The positive expression of PIK3 CA is closely correlated to the histological differentiation（P 〈0.01）, lymph node metastasis（P〈 0.05）. The frequency of PIK3 CA ‘hotspot＇ mutations, which are H1047 R, E542 K and E545 K, revealed no significant differences. Conclusion Endometriosis-associated ovarian carcinoma was negatively correlated with the mutation of PIK3 CA. However, the expression level of PIK3 CA was positively correlated with occurrence and pathogenesis of endometriosis-associated ovarian carcinoma.
