福建地区猪瘟病毒流行株基因分型及E2基因遗传变异分析 被引量:10

Genetic variation of E2 gene and analysis of genotyping classical swine fever virus in Fujian areas

摘要 为了解福建地区2013—2014年猪瘟病毒(CSFV)流行毒株的分子生物学特性与变异规律,采集了福建地区多个猪场的40份疑似猪瘟样品,应用RT-PCR技术对E2基因的主要抗原编码区进行扩增及序列分析,其中31份为猪瘟阳性。序列分析结果表明,31株CSFV流行毒株之间核苷酸与氨基酸的同源性分别为78.5%~100.0%和82.2%~100.0%,与国内外分离株的核苷酸同源性为81.1%~98.9%,氨基酸同源性为79.8%~95.6%;遗传进化树分析表明,31株CSFV属于1.1和2.1亚群,其中23株为1.1亚型,8株为2.1b和2.1c亚群,没有发现2.2和2.3亚群毒株。氨基酸序列分析表明,流行株在免疫逃逸相关位点705,713,729和734位氨基酸发生变异,FJ02毒株B细胞表位关键位点771位氨基酸发生变异,表明福建地区CSFV流行毒株呈现遗传变异多样性。 To understand classical swine fever virus (CSFV) molecular biology characteristics and variation regularity of epidemic strains in Fujian province from 2013 to 2014. The samples were collected from suspected CSF pigs from large-scale pig farms in Fujian province. The E2 genes were sequenced and analyzed. The results showed that the homologies of nucleotide and amino acid sequences of the thirty-one strains were 78.5%-100.0% and 82.2 ~/^-100. 0% ,respectively;the nu- cleotide and amino acid homology of E2 main antigen coding regions between 31 epidemic strain and different reference CSFV isolates were 81. 1%-98.9% and 79.8%-95.6%, respectively. The phylogenetic tree analysis showed that 31 isolated strains belonged to subgroup 1.1 and 2.1. Of the total,23 isolates belonged to subgroup 1.1,and eight isolates were subgroup 2. lb and 2. lc. Some variations in epidemic strains occurred at the sites (aa705,aa713,aa729 and aa734) related to immune evasion and a variation (aa771) at the critical site of B cell epitope of strain FJ02. These results showed that CSFV epidemic strains has undergone a rapid genetic diversity in Fujian prov- ince.
