摘要
银叶真藓(Bryum argenteum Hedw.)是藓类植物真藓科真藓属的模式种。世界广布种。该研究以银叶真藓为试材,采用流式细胞术,首先选用同属于藓类植物而且已公布基因组大小的小立碗藓(Physcomitrella patens)作为内参,估算得到银叶真藓基因组大小为568~589 Mb。鉴于参照的小立碗藓基因组大小为487 Mb是测序结果,可能在实际反映其基因组大小上有偏差,所以作者又选取了既有测序结果(125 Mb)又有流式细胞术估算基因组大小数据(157 Mb)的拟南芥(Arabidopsis thaliana)作为内参,对银叶真藓基因组大小进行了再次估算。结果表明,银叶真藓的基因组大小为728.09±77.84 Mb,1CDNA含量约为0.75 pg。该研究所得数据有助于银叶真藓基因组测序的研究。
Bryum argenteum Hedw., which belongs to Bryum in Bryaceae, is type specie of moss. The world has cosmopolitan species. In this study, the author firstly selected Physcomitrella patens as an internal reference, which was the homogeneous moss and the genome size was announced, using B. argenteum as a testing material and flow cytometry measurements. The estimated genome size of B. argenteum was 568~589 Mb. In view of the referential genome size of P. patens obtained by genome sequencing was 487 Mb, the genome size might be reflected with deviation in fact. Therefore, the author secondly selected the Arabidopsis thaliana(L.) Heynh. ecotype Columbia as an internal reference, which had both the sequencing results(125 Mb) and genome size data(157 Mb) estimated by the flow cytometry. And the author estimated the genome size of B. argenteum again. Finally, the result indicated that the genome size of B. argenteum was 728.09±77.84 Mb and 1C DNA was about 0.75 pg. The data obtained in this study is helpful to the research on genome sequencing of B. argenteum.
