摘要
目的对藏药镰形棘豆及其易混伪品进行分子鉴定,以确保该药材的质量及临床疗效。方法提取样本基因组DNA,对核基因ITS2片段进行PCR扩增并测序,并对所得序列进行同源性比较,利用MEGA软件进行相关数据分析,并构建基于ITS2序列的邻接(NJ)树。结果镰形棘豆物种种内K2P遗传距离为0,与其他混伪品的K2P遗传距离分布于0.068~0.084,构建的NJ树表明ITS2序列能够区分镰形棘豆与其他易混伪品。结论 ITS2序列作为DNA条形码可以方便快捷地鉴别镰形棘豆及其易混伪品,为其种质资源鉴定及临床安全用药提供了重要分子依据。
Objective To identify Oxytropis falcate Bunge from its adulterants; To secure the quality and clinical efficacy of Oxytropis falcate Bunge. Methods Genomic DNA was extracted. ITS2 fragment of nuclear gene was amplified and sequenced. The homology of the sequences was analyzed, and the related data were analyzed by software MEGA. The NJ(neighbor-joining) tree was constructed based on ITS2 sequence. Results The intraspecific genetic distance of Oxytropis falcate Bunge(K2P distance) was 0, and the interspecific genetic distance(K2P distance) between Oxytropis falcate Bunge and its adulterants ranged among 0.068-0.084. The constructed NJ tree showed that ITS2 could identify Oxytropis falcate Bunge from its adulterants correctly. Conclusion ITS2 barcode can be used to identify Oxytropis falcate Bunge from its adulterants effectively, and this study provides important molecular evidence for the identification of Oxytropis falcate Bunge and safety in its clinical use.
