摘要
目的研究拳卷地钱总黄酮对四氯化碳(CCl_4)致急性肝损伤大鼠的保护作用,并探索作用机制。方法采用腹腔注射10%(V/V)CCl_4花生油(5ml·kg^(-1))建立急性肝损伤大鼠模型,分光光度法检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、超氧化物歧化酶(SOD)、丙二醛(MDA)和谷胱甘肽过氧化物酶(GSH-Px)含量或活性,酶联免疫吸附试验(ELISA)检测肝组织中肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)和白介素-6(IL-6)水平,观察肝组织病理变化。结果与模型对照组比较,拳卷地钱总黄酮组(200 mg·kg^(-1)·d^(-1))能显著降低血清中ALT和AST活性,并能显著升高SOD活性,减少MDA含量,GSH-Px活性无明显变化,显著抑制肝组织中TNF-α、IL-1β和IL-6表达水平,改善肝组织病理变化。结论拳卷地钱总黄酮对CCl_4致急性肝损伤大鼠具有保护作用,其作用机制可能与抑制脂质过氧化、抗炎作用有关。
Objective To investigate the protective effect and related mechanism of total flavonoids from Marchantia convoluta on acute hepatic injury induced by carbon tetrachloride（ CCl4） in rats. Methods Using intraperitoneal injection of 10% CCl4 peanut oil to induce acute liver injury model in rats,spectrophotometry to detect the serum content or activity of alanine aminotransferase（ ALT）,aspartate aminotransferase（ AST）,malondialdehyde（ MDA）,superoxide dismutase（ SOD）,glutathione peroxidase（ GSH-PX）,enzyme-linked immunosorbent assay（ ELISA） to analyze the levels in liver tissue of tumor necrosis factor-α（ TNF-α）,interleukin-1β（ IL-1β） and interleukin-6（ IL-6）,and observing the hepatic pathological changes. Results Compared with the model group,the total flavonoids from Marchantia convoluta（ 200 mg·kg^-1·d^-1） could significantly reduce the serum activity of ALT and AST,the content of MDA,and increase the activity of SOD,but the activity of GSH-Px changed in significantly. It could also significantly inhibit the the levels in liver tissue of tumor necrosis factor-α（ TNF-α）,interleukin-1β（ IL-1β） and interleukin-6（ IL-6）,and improve liver pathological changes. Conclusion The total flavonoids from Marchantia convoluta had the protective effect on acute hepatic injury induced by CCl4 in rats,the mechanism maybe related with anti-inflammatory.
