柴胡皂苷d联合黄芩苷对脑缺血再灌注损伤大鼠抗氧化酶的影响 被引量:19

Effects of Saikosaponin d Combined with Baicalin on Antioxidant Enzymes in Rat with Cerebral Ischemia-Reperfusion Injury

摘要 目的探讨柴胡皂苷d联合黄芩苷对脑缺血/再灌注损伤大鼠抗氧化酶的作用。方法雄性SD大鼠60只,采用线栓法制备脑缺血再灌注模型,按随机数字表法分为正常对照组、假手术组、模型组、柴胡皂苷d组(70 mg·kg^(-1))、黄芩苷组(30 mg·kg^(-1))、柴胡皂苷d(70 mg·kg^(-1))+黄芩苷组(30 mg·kg^(-1)),缺血/再灌注后,每天腹腔注射一次,连续给药7d后,检测大鼠神经功能损伤评分,检测脑组织、血清中丙二醛(MDA)含量、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX))的活性。结果与假手术组比,模型组大鼠神经功能损伤评分显著升高,差异有统计学意义(P<0.01);药物组神经功能损伤评分显著降低,差异有统计学意义(P<0.01)。与假手术组相比,模型组脑组织、血清中MDA含量增加,差异有统计学意义(P<0.01),CAT、SOD、GSH-PX活性降低,差异有统计学意义(P<0.01);药物组脑组织、血清中MDA含量降低,差异有统计学意义(P<0.05),CAT、SOD、GSH-PX活性增强,差异有统计学意义(P<0.05)。柴胡皂苷d(70 mg·kg^(-1))+黄芩苷组(30 mg·kg^(-1))抗氧化酶作用显著。结论柴胡皂苷d联合黄芩苷对脑缺血再灌注损伤大鼠的保护作用可能通过抗氧化作用实现。 Objective To study the effects and mechanism of saikosaponin d combined with baicalin on antioxidant enzymes in rats with cerebral ischemia-reperfusion injury. Methods Sixty SD rats were ran- domly divided into the control group, sham group, model group, saikosaponin d group (70 mg/kg), baicalin group (30 mg/kg), and saikosaponin d (70 mg/kg) +baicalin group (30 mg/kg). Rats were pre- pared with cereral ischemia-reperfusion injury by suture method. After ischemia-reperfusion, they were given drugs once daily for seven consecutive days, then neurological deficits were determined. The content of malondialdehyde (MDA) in brain tissue and serum, and the activities of catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GSH-PX) were determined by the kits methods. Results Compared with sham group, the neurological scores significantly increased in model groups (P〈0.01), and decreased in drugs groups (P〈0.01). Compared with sham group, the content of MDA increased (P〈0.01), the activities of CAT, SOD, GSH-PX decreased (P〈0. 01) in brain and serum in model group with significant difference, and saikosaponin d+baicalin reduced the content of MDA (P〈 0.05) and elevated the activities of CAT, SOD and GSH-PX (P〈0.05) in brain and serum in saikosapo- nind and haicalin group. Saikosaponin d+haicalin had better effect on antioxidant enzymes. Conclusion The protection of saikosaponin d + baicalin against through its anti-oxidative effect. cerebral ischemia-reperfusion iniury might function
