摘要
对4个国家(地区)的13份西番莲种质用ITS、trnL-F、trnH-psbA等3种DNA条形码进行遗传多样性分析。结果表明,trnH-psbA条形码扩增出来的DNA序列的GC含量最低(27%),ITS条形码的DNA序列GC含量最高(63%);而trnL-F条形码扩增的DNA序列GC含量在不同种质间差异最大,在52.6%~62.3%之间。将3种DNA条形码序列在GeneBank中进行Blast比对,ITS和trnL-F序列基本与已登录种序列一致,达到77%以上,而trnH-psbA条形码序列与已登录种不一致。ITS条形码在序列间基因进化多样性分析显示,两两序列之间都具有差异;trnLF条形码序列的两两比较发现T1,T10,T11,T12,T13条形码序列之间无差异;trnH-psbA条形码序列的两两比较分析的差异最小,而H4与其他的序列差异较大,在13.3以上。从系统进化树来看,trnH-psbA条形码序列除了H4之外,其他样品聚为一类;trnL-F条形码序列中有5个不存在遗传差异;ITS条形码序列聚类的结果显示,每种西番莲均存在一定的遗传距离,这与基因多样化分析的结果一致。表明ITS在不同种质之间具有较高的多样性,可考虑应用于种质鉴定。
ITS barcode, trnL-F bareode, and trnH-psbA DNA barcode were used to analyze the genetic diversity of 13 Passiflora germplasms from four different countries or areas. The results showed that trnH-psbA barcode was with the lowest GC content (27%), ITS barcode with the highest (63 %), and trnL-F barcode with significantly different GC contents among germplasms (52.6%%62.3%). The sequences of 3 barcode was aligned by blast in GeneBank. All of ITS and trnL-F sequences were over 77% similar to the published accessions. The trnH-psbA barcode se- quences were unconformable with the accessions. The results of diversity analysis of gene evolution of ITS showed that there was difference between any two sequences. There was no evolutionary di- vergence among T1, T10, Tll and T12 in their trnL-F barcode diversity. The trnH-psbA barcodes showed the minimal diversity between any two germplasms. On the phylogenetic tree, all the sam- ples of trnH-psbA barcode were clustered into one group except H4, and there was no genetic difference among 5 of the trnL-F barcode sequences. ITS clustering showed genetic difference in some extent among all sequences, which accorded with the results of genetic diversity analysis. It was indicated that there was higher diversity among different Passiflora germplasms with their ITS bareode, so the ITS barcode could be used in germplasm identification.
