摘要
采集新疆苹果产区苹果树腐烂病标本,以组织分离法从标本中获得了5个菌株:PG576、PG431-2、PG534、PG650-1、PG787-1,通过室内致病性测定发现,5株菌均能够在健康枝条上致病且表现与田间相同的症状。经形态学特征结合多基因片段(ITS、NL、EF、ACT)的分子系统发育分析,明确PG576、PG431-2、PG534、PG650-1、PG787-1菌株为Cytospora schulzeri,首次发现该菌能够在新疆苹果树上造成危害。5株菌的最适生长条件为:PDA培养基,pH值6.0~7.0,28℃连续光照培养。
Pathogenic species of apple tree canker were collected from Xinjiang apple producing area and 5strains were obtained by tissue isolation method:PG576,PG431-2,PG534,PG650-1and PG787-1.The results showed that 5strains were able to cause disease in healthy branches and the same symptoms in the field.The molecular phylogenetic analysis based on the morophology characteristics and gene fragment(ITS,NL,EF,ACT)showed that Cytospora schulzeri can cause damage on apple tree.At the same time,the results showed that the optimum growth conditions of the 5strains were:PDA culture medium,pH value of 6.0-7.0,continuous illumination at 28 ℃.The research results have not only enriched the species of apple tree canker in Xinjiang,but also provided the theoretical basis for the investigating epidemiology of apple tree canker in the field.
