摘要
目的探讨参松养心胶囊和稳心颗粒对大鼠心肌纤维化的影响及其抗心律失常的机制。方法选取自发性高血压大鼠(SHR)40只随机分为4组,每组10只,对照组(C组,正常饲料喂养),缬沙坦组(VAL组,缬沙坦胶囊内容物混合饲料喂养),参松养心胶囊组(SHEN组,参松养心胶囊内容物混合饲料喂养),稳心颗粒组(WEN组,稳心颗粒混合饲料喂养)。喂养2个月后,进行心室内快速刺激,记录心律失常发生。之后处死大鼠,进行左心室组织切片,染色处理,观察各组切片纤维化程度,并采用免疫组化测定心肌细胞缝隙连接蛋白(Cx43)及基质金属蛋白酶-2(MMP-2)表达程度,进行比较。结果喂养2个月后,VAL组、SHEN组及WEN组心律失常诱发次数均较C组明显减少(P<0.05);图像定量分析显示VAL组、SHEN组及WEN组Cx43增高,MMP-2和组织金属蛋白酶抑制因子-2(TIMP-2)和荧光信号的面积百分比及积分光密度值较C组明显降低(P<0.05),且排列相对清晰。结论参松养心胶囊及稳心颗粒具有改善心肌重构的作用,能延缓心肌纤维化发展,降低致命性心律失常的发生率;其机制可能与增加Cx43及MMP-2的表达有关。
Objective To study the effect of Shensong Yangxin capsule (SYC) and Wenxin granule (WXG) on myocardial fibrosis in rats and its antiarrhythmic mechanism.Methods Forty spontaneously hypertensive rats (SHR) were randomly divided into 4 groups: control group (n = 10) fed with normal feed, valsartan group ( n = 10) fed with valsartan capsule content mixed feed, SYC group (n = 10) fed with SYC contents mixed feed. WXG group ( n = 10) fed with WXG contents mixed feed for 2 months. After 2 months of feeding,rapid ventricular stimulation was performed,and the occurrence of arrhythmia was recorded.After the rats were killed,the left ventricular tissue sections were stained, the degree of fibrosis was observed.The expression of connexin 43 (Cx43), matrix met-alloproteinase -2 (MMP-2) and tissue inhibitor of matrix metalloproteinase -2 (TIMP -2) in cardiac muscle cells was measured by immunohistochemistry. Results After feeding for 2 months, the occurrence of arrhythmia in valsartan group,SYC group,and WXG group were significantly less than that in control group ( P 〈0.05). Quantitative image analysis showed that the expressions of Cx43 in valsartan group,SYC group,and WXG group were higher than that in control group ( P 〈0 . 0 5 ),while the expressions of MMP-2 and TIMP-2 in valsartan group,SYC group,and WXG group were lower than that in control group ( P 〈0.05). Conclusion SYC and WXG has the effects on myocardial remodeling, can delay the development of myocardial fibrosis, thereby reducing the incidence of fatal arrhythmia.The mechanism may be related to the increased expression of Cx43 and MMP-2.
