摘要
[目的]优选海芋Alocasia macrorrhiza(Lour.)Schott多糖的最佳提取工艺,并分析其单糖组成。[方法]通过L9(34)正交试验设计,以多糖提取率为评价指标,优选海芋多糖的最佳提取工艺;多糖经酸水解、PMP衍生化,高效液相色谱分析其单糖组成。[结果]海芋多糖的最佳提取工艺:95℃,料液比1:20,回流提取3次,每次2h;在此条件下,多糖提取率为11.20%;PMP衍生化高效液相色谱法分析得海芋多糖主要由甘露糖、半乳糖醛酸、葡萄糖、半乳糖、阿拉伯糖等单糖组成。[结论]本实验成功建立简便、稳定可行的海芋多糖提取工艺,并对其单糖组成进行了研究,为合理地开发海芋资源提供了一定理论依据。
[Objective]To optimize the extraction process for polysaccharide from rhizome of Alocasia macrorrhiza and analyse the monosaccharide composition. [Methods]The rate of polysaccharide from rhizome ofAIocasia macrorrhiza was used as the index by L9(3^4) orthogonal design to optimize the best technique. Pre-column Derivatization HPLC was used to analyse the monosaccharide composition of polysaccharide from rhizome of Alocosia macrorrhizo, which was hydrolyzed by 3.0mo]/L HC1. [Results]The optimal technological condition of extraction process for polysaccharide from rhizome of Alocasia macrorrhiza was as followed: the extraction temprature was 95℃, material-liquid ratio was 1:20, the extraction time was 2h for 3 times. With the optimal extraction condition, the yield of polysaccharide from rhizome of Alocosia macrorrhiza was 11.20%; in addition, polysaceharide from rhizome of AIocasia macrorrhiza was composed by D(+)-mannose, D-Galacturonic acid, D(+)-glucose, D(+)-galactose, L(+)-arabinose. [Conclusion]The tests establish a steady, convenient extraction technology of polysaccharide from rhizome of Alocasia macrorrhiza and analyse the monosaccharide composition of polysaccharide, which provides a theoretical basis for the reasonable development of A locasia macrorrhiza.
