摘要
利用正交试验优化平菇下脚料中麦角硫因的回流提取工艺。通过高效液相色谱法,Kromasil100-5NH_2柱(250mm×4.60 mm,5μm),流动相乙腈-5 mmol/L醋酸铵(80∶20,体积比),254 nm下进行检测,建立麦角硫因的含量测定方法,以平菇下脚料中麦角硫因的含量及得率为评价指标。在单因素试验的基础上采用正交设计方法 L_9(3~4)考察提取时间、提取次数和料液比对麦角硫因提取效果的影响。结果表明平菇下脚料中麦角硫因的最佳提取工艺条件为:提取溶剂10%乙醇,料液比1∶6(g/m L),提取时间为2 h,提取2次。通过验证试验,麦角硫因的总提取率可达87.45%,含量可达0.5%。
The extraction technique for ergothion eine from mushroom scraps of Pleurotus ostreatus (Ft.) Kummer was optimized.The analysis were performed on Kromasil 100-5NH2 column with mobile phase consisted of acetonitrile-5 mmol/L ammonium acetate solution (80 : 20, volume ratio). The detected wavelength was set at 254 nm. The content and the extraction yield of ergothioneine were used as index to evaluate the technologies based on single factor test, in which three factors were considered the concentration of extraction time, extraction times and solid-liquid ratio by orthogonal design. The best extraction conditions were as follows: ethanol concentration of 10 %, twice, extraction time of two hours and ratio of liquid to solid of 1 : 6 (g/mL). The optimized extraction rate could reach 87.45 %, the content of ergothioneine was 0.5 %.
