摘要
目的分析血清PTEN、COX2蛋白表达与大肠癌复发转移的相关性,为临床治疗提供参考。方法选择160例复发转移大肠癌患者作为复发转移组,100例同期健康体检者作为对照组,120例初发大肠癌患者作为初发大肠癌组,120例复发未转移大肠癌患者作为复发未转移组。比较分析各组患者的血清PTEN、CD105、VEGF、COX2、PI3K、AKT蛋白表达水平,以及基质金属蛋白酶MMP1、MMP3、MMP9及其抑制剂TIMP1、TIMP2、TIMP7水平。结果复发转移组患者的血清PTEN蛋白表达水平低于对照组、初发大肠癌组及复发未转移组(P﹤0.05),而CD105、VEGF、COX2、PI3K、AKT、基质金属蛋白酶及其抑制剂水平高于对照组、初发大肠癌组及复发未转移组(P﹤0.05)。血清PTEN水平与复发转移大肠癌患者的预后呈正相关(r=0.9076,P﹤0.05),血清COX2水平与复发转移大肠癌患者的预后呈负相关(r=-0.9022,P﹤0.05)。结论大肠癌复发转移与血清PTEN水平降低及COX2水平升高有关,血清PTEN和COX2有望成为判断大肠癌复发转移及预后的重要生物学指标。
Objective To explore the association between the expression of serum PTEN and COX2 and the recur- rence and metastasis of colon cancer. Method 160 cases of recurrent and metastatic colon cancer patients were enrolled in this study, in the meantime 100 healthy volunteers were recruited to serve as controls for another 120 cases of primary colon cancer, and 120 cases of non-metastatic recurrent colon cancer. The expression of serum PTEN, CD 105, VEGF, COX2, PI3K, and AKT protein in each group were assayed, and the level of MMP1, MMP3, MMP9 with respective in- hibitors of TIMP 1, TIMP2, TIMP7 were detected. Result The serum PTEN was decreased in recurrent metastatic colon cancer patients compared with the other three groups （P〈0.05）, while the serum CD105, VEGF, COX2, PI3K, AKT, all MMP and responding inhibitors were higher in recurrent and metastatic cancer group than those in control group, primary cancer group and non-metastatic recurrent cancer group （P〈0.05）. It was obvious that PTEN was positively correlated with patients＇ prognosis （r=0.9076, P〈0.05）, while a negative correlation was observed with COX2 （r=-0.0922, P〈0.05）. Conclusion The recurrence and metastasis of colon cancer is related to the increased serum COX2 and decreased serum PTEN, which is possibly useful in predicting the recurrence, metastasis and prognosis of colon cancer.
