摘要
目的:建立3种大鼠乙酸型胃溃疡模型并进行评价。方法:将72只健康Wistar大鼠分为正常组、乙酸注射低、中、高3个剂量组(0.01、0.03、0.05m L)、乙酸滤纸贴敷组、乙酸直接灼烧组等6个组。采用乙酸注射、乙酸滤纸贴敷和乙酸直接灼烧法建立三种乙酸型胃溃疡大鼠模型。各组大鼠处死后通过肉眼观察溃疡点状,采用图像分析软件计算image溃疡面积;检测血清MDA、SOD和NO的含量。结果:乙酸注射低、中、高剂量组溃疡面均较小,但中剂量组﹑高剂量组与正常组比较均有统计学差异(P<0.05、P<0.01);乙酸滤纸贴敷组和乙酸直接灼烧组溃疡面较大,与正常组比较均有统计学差异(P<0.01)。与正常组比较乙酸注射中剂量组、乙酸滤纸贴敷组、乙酸直接灼烧组的血清MDA含量均有统计学差异(P<0.05、P<0.01);乙酸直接灼烧组血清SOD含量与正常组比较有统计学差异(P<0.05);乙酸注射中剂量组、乙酸滤纸贴敷组和乙酸直接灼烧组血清NO水平高于正常组,均有统计学差异(P<0.05、P<0.01);与正常组相比,乙酸注射高剂量组、乙酸滤纸贴敷组和乙酸直接灼烧组的EGF的浓度,均有统计学差异(P<0.05、P<0.01)。结论:乙酸直接灼烧法导致的胃溃疡模型最为理想,适用于慢性胃溃疡大鼠模型的建立。
Objective Establish three kinds of acetic acid type gastric ulcer model in rats and evaluated. Method Will be divid- ing, sixgroups. After building 14, groups of rats with anesthesia, open, blood, stomach, macroscopic observation point of ulcer after taking pictures, measured by image analysis software Imagej ulcer area; Detection of serum MDA, SOD and the content of NOS and the concentration of EGF. Results Acetic acid injection ulcer surface low, medium and high dose group were smaller, but the medium dose group and high dose group compared with normal group, were statistically significant (P 〈0. 01, P 〈0. 05) ; The group of acetic acid and the direct burning group of acetic area compared with normal group, there were statistical differences (P 〈 0. 01 ) . compared with normal group, acetic acid injection dose group, acetate filter paper sticking group, direct calcination acetate group of serum MDA content were statistically significant (P 〈 0. 05, P 〈 0. 01 ) . compared with normal group, the serum SOD content of acetic acid directly burning group was statistically different (P 〈 0. 05) . compared with normal group, acetic acid injection dose group, acetate filter paper sticking group, direct calcination acetate group of serum NOS content were statistically significant (P 〈 0. 05, P 〈 0. 01 ) . compared with normal group, the acetic acid injection of high dose group, the acetate filter paper sticking group and the concentration of acetic acid direct calcination group of EGF, were statistically significant ( P 〈 0. 05, P 〈 0. 01 ). Conclusion The ideal of the gastric ulcer model caused by the direct burning method of acetic acid is suitable for the establishment of rat model of the chronic gastric ulcer.
