小剂量玉米赤霉烯酮损害雄性小鼠生殖能力的研究 被引量:4

摘要 目的评估小剂量(低于无明显作用水平的浓度)玉米赤霉烯酮(zearalenone,ZEA)暴露后小鼠精子发生及精液质量的变化趋势,探讨小剂量ZEA暴露对雄性小鼠生殖能力的影响。方法 90只出生后21 d的雄性CD-1小鼠按随机数字表法分成3组,分别暴露于0、20、40μg/(kg·d)ZEA,处理14、28、42 d后,采用计算机辅助精液质量分析观察精子活力参数,吉姆萨染色观测精子畸形率,测量睾丸体积及质量,免疫组化染色检测生精细胞计数。结果处理42 d后,20、40μg/(kg·d)组精子活动率低于0μg/(kg·d)组[(82.80±4.34)%、(74.90±6.70)%vs(93.20±1.66)%,P=0.004、0.002];20、40μg/(kg·d)组小鼠畸形精子率高于0μg/(kg·d)组[(29.40±1.09)%、(40.80±6.17)%vs(19.40±0.45)%,P<0.001,P=0.002];20、40μg/(kg·d)组小鼠的单位面积生精细胞计数低于0μg/(kg·d)组(2 422.7±206.1、1 855.8±105.3 vs 3 189.8±306.0,P=0.013、0.001)。结论小剂量ZEA的暴露能够降低雄性小鼠的精子发生及精液质量,损害其生殖能力。 Objective To investigate the changes of spermatogenesis and semen quality in mice exposed to low-dose (below the no-observed effect level) zearalenone (ZEA) and assess the effect of such exposures on the reproductive capacity of male mice. Methods Ninety male CD-I mice (21 days old) were randomized into 3 groups for exposure to intragastrieally administered ZEA at 0, 20 and 40 μg/( kg ·d) for 14, 28 or 42 d. After the exposures, the sperm motility of the mice was assessed using the computer-aided semen analysis system, and sperm deformity was detected with Giemsa staining. The weight and volume of the testis were measured, and the count of spermatogenic cells per unit area was evaluated with immunohistochemical staining. Results Exposure to ZEA for 42 d caused significantly lowered sperm motility in 20 and 40 μg/( kg · d) dose groups in comparison with the unexposed group [ ( 82.80 ± 4.34 ) %, (74.90 ±6. 70)%, and (93. 20 ± 1. 66)%, respectively; P =0.004, 0.002]. Compared with the unexposed group, exposure to 20 and 40 μg/( kg ·d) ZEA resulted in significantly increased sperm deformity rate [(29.40±1.09)%, (40.80±6.17)% vs (19.40±0.45)%, P〈0.001, P=0.002] and reduced counts of spermatogenic cells per unit area (2 422.7 ± 206.1, 1 855.8± 105.3 vs 3 189.8 ± 306.0, P = 0. 013, 0. 001). Conclusion Low-dose ZEA exposure impairs spermatogenesis and semen quality and reproductive capacity in male mice.
