摘要
目的:研究秦七风湿方水提液和醇提液对巨噬细胞RAW264.7增殖和分泌一氧化氮(NO)的影响。方法:采用水煎煮和乙醇超声分别制备秦七风湿方水提液和醇提液,通过HPLC测定水提液和醇提液指标性成分含量;利用CCK-8研究水提液和醇提液对RAW264.7增殖的影响;通过Greiss法测定水提液和醇提液对巨噬细胞分泌NO的影响。结果:含量测定发现醇提液的指标性成分含量高于水提液,说明不同提取溶剂所得秦七风湿方指标性成分具有差异;水提液和醇提液对RAW264.7增殖的影响呈现浓度依赖性,具有促进和抑制的作用;水提液和醇提液对LPS诱导的巨噬细胞分泌NO具有显著的抑制作用。结论:通过研究发现秦七风湿方水提液和醇提液对RAW264.7增殖和分泌NO均具有一定的影响,呈现浓度依赖性,并且初步证实了水提液和醇提液均具有一定的抗炎活性,为将其发展为治疗RA的临床新药提供理论参考。
Objective： To study the effect of Qinqi Rheumatism formula aqueous extract and ethanol extract on the proliferation and production of nitric oxide （NO） with RAW264. 7 macrophage. Methods ： Qinqi Rheumatism formula aqueous extract and ethanol extract were prepared through aqueous decoction and ethanol ultrasonic respectively, the index components of aqueous extract and ethanol extract were detected by HPLC. The effect of aqueous extract and ethanol extract on the prolifera- tion of RAW264. 7 was evaluated using the CCK-8 assay. The effect of aqueous extract and ethanol extract on the secretion of NO was measured by the Griess reaction method. Results ： The index components of ethanol extract were higher than those of a- queous extract, which showed the differences of the index components of Qinqi Rheumatism formula prepared by different ex- traction solvents. The aqueous extract and ethanol extract promoted or inhibited the proliferation of RAW264. 7 in a dose-de- pendent manner. The inhibitory effect of the aqueous and ethanol extract in the production of NO in LPS-stimulated RAW264. 7 was found. Conclusion ： The effects of the aqueous extract and ethanol extract of Qinqi Rheumatism formula on the proliferation and secretion of NO in RAW264. 7 was found in a dose-dependent manner, and the anti-inflammatory activity of aqueous ex- tract and ethanol extract was confirmed preliminarily. This study will provide theoretical basis for developing Qinqi Rheumatism formula as a new drug for the treatment of RA.
