摘要
应用荧光影像系统检测了原代培养海马神经元内钙离子的变化情况 ,并分析了皮质酮、肿瘤坏死因子(TNF α)对谷氨酸引起的海马神经元内钙升高的调节作用及其皮质酮对TNF α作用的调节。结果显示 :(1)使用不同浓度的皮质酮处理海马神经元 4 8h后 ,观察到 10 6 mol/L和 10 7mol/L的皮质酮可诱导海马神经元静息钙浓度升高 ,但 10 8mol/L和 10 9mol/L的皮质酮对海马神经元内钙无影响。 (2 ) 10 6 mol/L皮质酮处理海马神经元 4 8h后 ,对谷氨酸诱导的海马神经元内钙升高无调节作用。 (3)使用 10 0ngTNF α处理海马神经元 4 8h后 ,既可诱导海马神经元静息钙升高 ,也可抑制谷氨酸引起的海马神经元内钙升高。 (4)皮质酮可逆转TNF α对海马神经元静息钙浓度的调节作用 ,但对TNF α抑制谷氨酸升钙效应无影响。
The effect of corticosterone and TNF α on the change of cytosolic free calcium (\[Ca 2+ \] i ) was investigated in primary culture hippocampal neurons by using fluorescence image system. The main results were as follows: (1) When cells were preincubated at 37 ℃ for 48 h in the presence of various concentration of corticosterone, 10 6 mol/L or 10 7 mol/L corticosterone could induce \[Ca 2+ \] i rise in hippocampal neurons ( P <0.05). (2) When hippocampal neurons were preincubated with 10 6 mol/L corticosterone at 37 ℃ for 48 h, the \[Ca 2+ \] i rise induced by 10 3 mol/L glutamate was not influenced. (3) When hippocampal neurons were preincubated with 100 ng TNF α at 37 ℃ for 48 h, bath the rising of resting \[Ca 2+ \] i and an inhibitory effect on \[Ca 2+ \] i rise induced by 10 3 mol/L glutamate were observed in hippocampal neurons ( P <0.01). (4) When hippocampal neurons were preincubated with 10 6 molL corticosterone and 100 ng TNF α at 37 ℃ for 48 h, the effect of TNF α on resting [Ca 2+ ] i was reversed, but [Ca 2+ ] i rise induced by glutamate was not influenced.
