摘要
目的观察子痫前期胎盘组织中miR-155和炎症因子的表达情况,探讨该miR对子痫前期炎症应答的调控。方法收集2014年1月至2015年12月在深圳市罗湖区人民医院住院的子痫前期患者和同期住院正常晚期妊娠孕妇各20例,应用Real-time PCR检测胎盘组织中miR-155和白细胞介素-1β、白细胞介素-6、肿瘤坏死因子-α的表达水平;分离培养胎盘滋养层细胞,采用miR-155抑制剂(anti-miR-155)敲低miR-155表达,通过real-time PCR检测IL-1β、IL-6、TNF-α的表达变化情况。结果子痫前期组胎盘组织内miR-155和IL-1β、IL-6、TNF-α表达量较正常对照组明显升高,敲低miR-155后显著抑制IL-1β、IL-6、TNF-α的表达。结论 miR-155参与调控子痫前期滋养层细胞IL-1β、IL-6、TNF-α的表达,从而加剧子痫前期炎症反应。
Objective To investigate whether miR-155 and inflammatory factors were expressed in preeclamptic placentas and discuss the regulation effect of miR-155 on preeclampsia inflammatory response. Methods Twenty patients with preeclampsia and 20 cases of normal pregnant women(control group)were selected from the People's Hospital of Luohu District of Shenzhen city from January 2014 to December 2015.The expression of miR-155,IL-1β,IL-6 and TNF-α was determined by using real-time PCR.Trophoblast cells were separated and cultured,specific anti-miR-155 was used to knock down miR-155 expression,and the expression change of IL-1β,IL-6 and TNF-α were measured by using real-time PCR.Results Expression of miR-155,IL-1β,IL-6 and TNF-α was up-regulated in the preeclampsia placenta tissue.Knockdown of miR-155 significantly inhibited the expression of IL-1β,IL-6 and TNF-α.Conclusion The miR-155 is involved in regulating the expression of IL-1β,IL-6 and TNF-α in trophoblast cells and accelerate preeclampsia inflammation.
