摘要
目的:通过检测四逆散不同配伍中有效成分柴胡皂苷a在大鼠体内的药代动力学参数,发现药物之间的协同或拮抗作用,探讨四逆散配伍的合理性。方法:通过灌胃分别给予大鼠四逆散全方汤剂及7组拆方配伍汤剂,使用液相色谱仪检测柴胡皂苷a的血药浓度,利用计算机软件计算其药代动力学参数。结果:白芍与柴胡配伍明显提高了柴胡皂苷a在大鼠体内的最大浓度含量;炙甘草与柴胡配伍不仅提高了柴胡皂苷a在大鼠体内的最大浓度含量,还延长了柴胡皂苷a在大鼠体内的半衰期;枳实本身与柴胡配伍对大鼠体内柴胡皂苷a的药代动力学参数影响不明显,但多药合用则对柴胡皂苷a的半衰期有明显延长作用。综合比较各项参数,对四逆散中柴胡有效成分柴胡皂苷a的吸收率及血药浓度维持最为有益的配伍为四逆散全方。结论:四逆散全方结构严谨配伍精妙,白芍和炙甘草均对柴胡的功效具有积极作用,可促进其有效成分的吸收,并与枳实产生协同作用,共同维持柴胡中有效成分柴胡皂苷a的血药浓度,其组方配伍具有很高的科学性和合理性。
Objective:Through detecting saikosaponin a pharmacokinetic parameters of Sini San with the different compatibility in the rats,to find synergistic or antagonistic effect between drugs,to discuss rationality of Sini San compatibility. Methods:The whole decoction and seven compatibility decoction of Sini San were given to the rats through the intragastric administration. Blood concentrations of the saikosaponin a were detected by the liquid chromatograph. The pharmacokinetic parameters was calculated by using the computer software. Results:Compatibility of Chaihu and Baishao obviously improved the maximum concentration of saikosaponin a in the rats. Compatibility of Chaihu and Gancao not only improved the maximum concentration of saikosaponin a,but also extended the half-time of saikosaponin a in the rats. Compatibility of Chaihu and Zhishi had a little effect on the pharmacokinetic parameters of the saikosaponin a,whereas multi drug combination obviously extended the half-time of saikosaponin a. From the comprehensive comparison of various parameters,it concluded that Sini San whole decoction was the best compatibility. It was benefit for the absorption rate and the maintenance of blood concentrations of the saikosaponin a in Sini San. Conclusion:Sini San whole decoction had the strict structure and subtlety compatibility. Baishao and Gancao had a positive affect on the Chaihu efficacy that promoted the absorption of the active ingredient. Zhishi had synergistic reaction with them. They maintain the blood concentrations of the saikosaponin a that is the active ingredient in Chaihu. Its compatibility was high scientific and reasonable.
