摘要
目的探讨血浆微小RNA(miR)-424-5p在宫颈鳞癌中的表达情况及其与临床病理特征的关系并预测其可能的靶基因。方法收集200例宫颈鳞癌患者及100名健康女性的血浆样本,采用实时定量聚合酶链反应(RT-PCR)测定血浆miR-424-5p水平,分析血浆miR-424-5p水平与宫颈鳞癌临床病理因素的关系,并通过生物信息学分析,预测miR-424-5p可能的靶基因。结果宫颈鳞癌患者血浆miR-424-5p相对表达水平低于健康对照组(P=0.023);血浆miR-424-5p相对表达水平与宫颈鳞癌的临床分期有关(P<0.05);而与及肿瘤分化程度、淋巴结转移、年龄、绝经状况无相关性(P>0.05)。通过生物信息学分析,miR-424-5p预测的靶基因多数与细胞周期因子有关。结论 miR-424-5p有可能通过调节细胞周期因子在宫颈癌发生、发展中起作用。
Objective To investigate the expression of plasma miR-424-5p in cervical squamous cell carcinoma and its relationship with clinicopathological characteristics, and to predict its potential target genes. Methods The plasma samples of 200 patients with cervical squamous cell carcinoma and 100 healthy women were included. The real-time RT-PCR was used to determine plasma miR-424-Sp level. The relationship between plasma miR-424-Sp level and clinieopathological factors of cervical squamous cell carcinoma was analyzed. The potential target genes of miRNA-424-Sp were predicted by bioinformatics analysis. Results The relative expression level of plasma miR-424- 5p in patients with cervical squamous cell carcinoma was lower than that in the normal control group (P=0.023). The relative expression level of miR-424-Sp was correlated with the clinical stage of cervical squamous cell carcinoma (P〈 0.05), but was not correlated with tumor differentiation, lymph node metastasis, age and menopausal status (P〉0.05). Through hioinformatics analysis, the majority of the target genes predicted by miR-424-Sp was correlated with cell cycle factors. Conclusion MiR-424-Sp may play a role in the development and progression of cervical cancer by regulating cell cycle factors.
