摘要
按体重将80只SD大鼠随机分为对照组,低、中、高剂量组,每组雌雄各10只。持续静脉给药观察4周后处死,观察体重、脏器系数、血液及生化指标以及病理切片。与对照组比较,各组大鼠体重无显著性差异(p>0.05);高剂量组雄鼠脑、肺、胸腺脏器系数差异显著(p<0.05),雌鼠肝、卵巢脏器系数有差异显著(p<0.05);中高剂量组雌、雄鼠部分血液及生化指标差异显著(p<0.05);病理切片观察,各组大鼠肝脏均出现脂变,中高剂量组大鼠附睾、大肠出现炎症,其他器官未见明显病理改变。中高剂量组ATBC溶液对大鼠有一定毒性,参考低剂量组给药浓度,大鼠对其安全剂量为500 mg/kg体重/d。
80 SD experimental rats were randomly divided into control group, low, medium and high dose groups based on body weight, with 10 male and female rats in each group. These rats were put to death after 4 weeks of continuous intravenous administration, and we observed the body weight, organ coefficient, blood and biochemical indicators and histopathology. Compared with the control group, the body weight of 3 experimental groups had no significant difference (p 〉0.05); In high dose group, the brain, lung, thymus organ coefficient of male rats were significantly different from the control group (p 〈0.05), and the liver, ovary organ coefficient of female rats had significant difference from those of the control group (p 〈0.05); In middle and high dose group, the blood and biochemical indicator of female and male rats showed significant difference (p〈0.05). The pathological observation showed that lipid changes occurred in the liver of each group. The epididymis and large intestine inflammation were found in the middle and high dose group. No obvious pathological changes were found in other organs. Therefore, the study suggested that middle and high dose ATBC liquid had a certain toxicity. Referring to the low dose group administration, the security tolerance value of ATBC in rats was 500 mg/kg bw/day.
