摘要
目的探讨白细胞介素-10(IL-10)、白细胞介素-17(IL-17)水平在乙型肝炎病毒相关性肝癌中的表达及其临床价值。方法选取2013年8月-2016年9月医院乙型肝炎病毒相关性肝癌患者104例作为研究组及同期健康体检者104名为对照组,比较乙型肝炎病毒相关性肝癌患者肝癌组织和癌旁组织及健康人群正常肝组织IL-10、IL-17表达及甲胎蛋白含量等。结果 IL-10在肝癌组织、癌旁组织中mRNA水平表达分别为(0.472±0.050)Gapdh、(0.022±0.004)Gapdh,高于健康人群的(0.010±0.002)Gapdh,IL-17在肝癌组织、癌旁组织中mRNA水平表达分别为(0.620±.0.087)Gapdh、(0.024±0.005)Gapdh,高于健康人群的(0.011±0.004)Gapdh,肝癌组织高于癌旁组织,差异均有统计学意义(P<0.05),肝癌组织、癌旁组织中蛋白水平表达高于健康人群,肝癌组织高于癌旁组织,差异均有统计学意义(P<0.05);在肝癌细胞株BEL-7402、HepG2中的水平高于正常肝细胞HL-7702,BEL-7402高于HepG2,差异均有统计学意义(P<0.05);IL-10、IL-17在mRNA和蛋白水平表达与甲胎蛋白含量呈正相关性(P<0.05)。结论 IL-10、IL-17高表达与乙型肝炎病毒相关性肝癌发生发展具有密切相关性,加强二者检测与干预,有助于乙型肝炎病毒相关性肝癌的诊断与疾病治疗,改善预后。
OBJECTIVE To investigate the expression of IL-10 and IL-17 in hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC) and its clinical value.METHODS From Aug.2013 to Sep.2016, 104 patients with hepatitis B virus-related liver cancer were selected as research group, and 104 healthy subjects were selected as control group.The levels of IL-10, IL-17 and alpha-fetoprotein in normal liver tissue of patients with hepatocellular carcinoma (HCC) and adjacent tissues of patients with hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC) were measured and compared.RESULTS The mRNA levels of IL-10 in HCC tissues and adjacent tissues were (0.472±0.050)Gapdh and (0.022±0.004)Gapdh, which were higher than(0.010±0.002)Gapdh in healthy subjects, the mRNA levels of IL-17 in HCC tissues and adjacent tissues were (0.620±0.087)Gapdh and (0.024±0.005)Gapdh, which were higher than(0.011&#177;0.004)Gapdh in healthy subjects, and the expression of IL-10 and IL-17 were significantly higher in HCC tissues than those in adjacent tissues (P&lt;0.05).The protein expression was significantly higher in HCC tissues and adjacent tissues than that in healthy subjects (P&lt;0.05).The levels in hepatocarcinoma cell line BEL-7402 and HepG2 were higher than thoase in normal hepatocyte HL-7702 and BEL-7402, respectively (P〈0.05).The expression of IL-10 and IL-17 was positively correlated with the content of alpha-fetoprotein at mRNA and protein level (P〈0.05).CONCLUSION The high expression of IL-10 and IL-17 is closely related to the development and progression of hepatitis B virus-associated hepatocellular carcinoma.To strengthen the detection and intervention of IL-10 and IL-17 is helpful to the diagnosis and treatment of hepatitis B virus-related hepatocellular carcinoma and can improve the prognosis.
