摘要
目的观察电针对术后认知功能障碍大鼠认知功能及海马TNF-?和IL-1?表达的影响,探讨电针的作用机制。方法将90只健康SD雄性老龄大鼠,随机分为假手术组(C组),手术组(S组),电针组(Z组),每组30只。每组又分为术后1 d、术后3 d和术后7 d 3个亚组。采用Morris水迷宫检测认知功能,免疫组化法检测大鼠海马TNF-?和IL-1?的变化。结果与C组同时点比较,S组术后1 d、3 d、7 d找到平台的潜伏期延长,穿越平台次数减少(P<0.05),以术后3 d最为明显;与S组同时点比较,Z组术后1 d、3 d、7 d找到平台的潜伏期缩短,穿越平台次数增多(P<0.05)。与C组同时间点比较,S组TNF-?和IL-1?术后阳性表达数量增加(P<0.05);与S组同时间点比较,Z组各亚组阳性表达数量降低(P<0.05)。结论电针调节能改善肝左叶切除大鼠术后学习和记忆能力,可能与抑制海马TNF-?和IL-1?的过度表达有关。
Objective To observe the effect of electroacupuncture (EA) on the cognition and hippocampal tumor necrosis factor (TNF)-α and interleukin (IL)-1β) expressions in rats with postoperative cognitive dysfunction (POCD), and discuss the action mechanism of EA. Method Ninety healthy male senile Sprague-Dawley (SD) rats were randomized into a sham operation group (group C), an operation group (group S) and an EA group (group Z), 30 rats in each group. Each group was then further divided into 3 subgroups: postoperative day 1, postoperative day 3 and postoperative day 7. The Morris water maze was adopted to evaluate the cognitive function, and immunochemistry method was used to detect the changes of TNF-α and IL- 1β. Result Compared with group C at the same time points, the latent period was significantly longer and the crossing times were lower in group S on postoperative day 1, day 3 and day 7 (P〈0.05), and the differences were most significant on postoperative day 3; compared with group S at the same time points, the latent period was significantly shorter and the crossing times were higher in group Z on postoperative day 1, day 3 and day 7 (P〈0.05). Compared with group C at the same time point, the positive expressions of TNF-α and IL-1β were significantly higher in group S (P〈0.05); compared with group S at the same time points, the positive expressions were significantly lower in each subgroup of group Z (P〈0.05). Conclusion EA can improve the learning and memory of rats after left lateral segmentectomy of the liver, which is possibly related to the inhibition of the over-expressions of TNF-α and IL-1β.
