中性粒细胞弹性蛋白酶抑制剂对急性百草枯中毒大鼠治疗作用的实验研究 被引量:2

Experimental study on the therapeutic effect of neutrophil elastase inhibitor on acute paraquat poisoning rats

摘要 目的探讨中性粒细胞弹性蛋白酶抑制剂(NEI)对急性百草枯中毒大鼠的治疗作用。方法 96只健康SPF级Wistar大鼠,随机分为对照组、模型组、治疗组,每组32只。以50mg/kg 20%百草枯水剂溶液一次性灌胃大鼠建立急性百草枯中毒动物模型,治疗组给予西维来司钠8mg/kg iv bid×3d。密切观察各组大鼠行为学改变,于染毒后第1、3、7、21天处死大鼠,采用ELISA法检测大鼠血清中血清白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)、转化生长因子-β1(TGF-β1)和中性粒细胞弹性蛋白酶(NE)浓度水平。结果与对照组相比,模型组各时间点血清IL-8、TNF-α、TGF-β1、NE水平较对照组均明显升高(P<0.05)。与模型组相比,治疗组在染毒后各时间点血清IL-8及TNF-α水平明显降低(P<0.05),血清TGF-β1水平第1、3天明显降低(P<0.05),血清NE第1、3、7天浓度水平较模型组明显下降(P<0.05)。结论 NEI对急性百草枯中毒大鼠有一定治疗作用,其机制可能与该药的抗炎作用有关。 Objective To explore the therapeutic effect of neutrophil elastase inhibitor in acute paraquat poisoning rats.Methods A total of 96 SPF Wistar rats were randomly divided into three experimental groups:control group,model group and therapy group and there were 32 rats in each group,Paraquat was given to rats by oral gavage at the dose of 50mg/kg.Sivelestat sodium was intraperitoneal injected to therapy groups' rats at the dose of 8mg/kg twice a day for three consecutive days;closely observe changes of behavior in rats.All rats were killed on the 1st,3rd,7th and 21 st day after the exposure.The levels of interleukin-8(IL-8),tumor necrosis factor-α(TNF-α),transforming growth factor-β1(TGF-β1)and neutrophil elastase(NE)in serum were detected by ELISA. Results Compared with the control group,the levels of IL-8,TNF-α,TGF-β1and NE in serum increased significantly in model group at different time points after administration(P〈0.05),Compared with the model group,the levels of IL-8and TNF-αin serum decreased significantly in therapy group(P〈0.05);the levels of TGF-β1in serum decreased significantly therapy group on the 1st and the 3rd day(P〈0.05),similar reductions also occurred in serum NE levels,except for day 21(P〈0.05). Conclusion NEI has therapeutic effect on acute paraquat poisoning rats,the mechanisms may be associated with antiinflammatory effects.
