摘要
为比较两种商品化赤羽病抗体ELISA试剂盒的敏感性和特异性,以及ELISA和微量血清中和试验(SNT)的符合率,采用两种ELISA试剂盒和血清中和试验,对690份澳大利亚纯种荷斯坦奶牛血清进行牛赤羽病检测。试验结果显示:法国某公司生产的赤羽病ELISA试剂盒敏感性为93.42%,特异性为81.23%,与SNT间的Kappa值为0.615,为高度符合;日本某公司生产的赤羽病ELISA试剂盒敏感性为39.47%,特异性为96.1%,与SNT间的Kappa值为0.427,为中度符合。比较结果表明:日本某公司生产的试剂盒敏感性较低,容易漏检,不适合用于牛赤羽病初筛;在出入境检疫中,可以采用高通量、半自动化、敏感性高的ELISA方法,对血清样本进行筛选,再采用特异性强的SNT试验进行辅助验证。
To compare the 2 commercial ELISA kits from the aspects of the diagnostic sensitivity（DSe）,the diagnostic specificity（DSp）,and the coincidence rate of ELISA and serum neutralization test（SNT）,the Akabane disease detection was carried out towards 690 serum samples of cattle imported from Australia,by two ELISA kits and serum neutralization test. The results showed that the DSe of the ELISA kit from a French company was 93.42%,the DSp was 81.23% and the Kappa with SNT was 0.615,which was consistent highly. The DSe of the ELISA kit from a Japan company was 39.47%,the DSp was 96.1% and the Kappa with SNT was 0.427,which was consistent moderately. The comparative results showed that the DSe of kit produced by Japanese company was low,and it was easy to miss detection,which was unsuited for Akabane disease detection. In the entry and exit quarantine,a high throughput,semi-automated and highly sensitive ELISA method could be used to screen the serum samples,and the SNT test with high DSp was carried out to assist the verification.
