摘要
阿尔茨海默病(AD)是一种以智能减退为主要特征的中枢神经系统疾病。AD危害极大,但病因复杂,病理不明,治疗方法不足。合适的AD鼠模型不仅能为富有科学价值的研究提供良好的实验研究材料,对于明确AD的病因、发病机制及在此基础上探索AD的预防、诊断及治疗等各方面也均发挥关键的作用。根据AD发病机制的假说:脑衰老、β淀粉样蛋白神经毒性学说、Tau蛋白异常磷酸化学说、铝中毒学说、胆碱能障碍学说及遗传基因突变学说等,AD鼠模型经历了从单一假说基础上的单一因素到多假说多因素造模的发展。AD模型鼠的发展为AD病理和诊治研究提供了研究的手段和途径。
Alzheimer's disease(AD) is a neurodegenerative disorder characterized by the progressive decline of intelligence. Although AD brings great harm to the patients, its pathology is complicated and elusive, and the current treatment is insufficient. The proper AD rodent models would provide valuable animal subjects for scientific research of AD. Based on the assumed etiologic and pathogenic factors of AD including brain aging, the neurotoxic β-amyloid, the abnormal phosphoryla- tion of Tau protein, the poisoning aluminum, the eholinergie disorder, and genetic mutation, the AD rodent models have developed from the single factor to multi-factors, which provides research means and approaches for the study of the pathology, diagnosis and treatment of AD.
