摘要
目的探讨三种不同消毒方法对电纺聚乳酸(PLA)支架性能和生物相容性的影响。方法采用红外光谱、X射线衍射、热重分析、四甲基偶氮唑盐(MTT)比色法、水接触角和扫描电镜检测经三种方法消毒后电纺PLA支架理化性能及其生物相容性,同时对比分析环氧乙烷改良法灭菌后支架的生物相容性。结果三种不同方法消毒后电纺PLA支架物理性能都有不同程度的改变,其中环氧乙烷组支架纤维融合最严重,乙醇组支架亲水性改善最为明显;MTT结果表明乙醇组支架上细胞增殖能力最强;环氧乙烷改良法灭菌后支架材料的生物相容性有明显提高。结论三种消毒方法均可用于电纺PLA材料的消毒,但乙醇浸泡法对电纺支架材料的组织工程学应用影响较小。
OBJECTIVE To investigate the effect of three different kinds of sterilization methods on the properties and biocompatibility of electrospun polylactic acid(PLA)scaffolds.METHODS After disinfection with the three methods,the properties and biocompatibility of electrospun PLA scaffolds were detected by infrared spectroscopy,X ray diffraction,thermogravimetric analysis,MTT,water contact angle and scanning electron microscopy.Meanwhile,the biocompatibility of electrospun PLA scaffolds was compared and analyzed after improved ethylene oxide sterilization.RESULTS Through the three methods,the physical properties of electrospun PLA scaffolds displayed different degrees of changes after sterilization.The ethylene oxide group had the most serious fiber fusion,while the hydrophilicity of the ethanol group improved most significantly.MTT showed that the ethanol group had the strongest cell proliferation on scaffolds,and the biocompatibility of scaffolds was obviously enhanced after improved ethylene oxide sterilization.CONCLUSIONAll of the three disinfection methods can be used to sterilize electrospun PLA materials,however the ethanol immersion method has comparatively less effect on the application of the electrospun scaffolds in tissue engineering.
