摘要
通过对雌三醇分子结构的改造,制备了雌三醇半抗原和人工抗原,通过对动物进行免疫得到雌三醇的单克隆抗体,该抗体灵敏度为0.05μg/L,对雌二醇和雌酮的交叉反应率小于10.00%。基于该抗体建立了雌三醇间接竞争酶联免疫检测方法,该方法检测环境水样品中雌三醇的检测限为0.25μg/L,回收率为72.00%~108.00%,变异系数小于15.00%。
By transforming the molecular structure of the estriol,we obtained the hapten and artificial antigens. The monoclonal antibodies of estriol were obtained by immunizing animals. The sensitivity of the antibody was 0.05 μg/L,and the cross-reaction rate to estradiol and estrone were less than 10. 00%. An indirect competitive enzyme immunoassay was constructed based on the antibody for the quantitative analysis of estriol. The results showed that the limit of detection of environmental water sample was 0. 25 μg/L. The range of average recovery rate was 72. 00% ~ 108. 00%,and the coefficients of variation were less than 15. 00%.
