摘要
目的 :观察不同剂量川楝子长期给药对大鼠肝肾的损伤以及炒制对川楝子肝肾毒性的影响。方法 :将63只健康wistar雄性大鼠,按照随机数字表法分为对照组、生川楝子高剂量组(10 g·kg^-1·d^-1、生川楝子中剂量组(5 g·kg^-1·d^-1、生川楝子低剂量组(2.5 g·kg^-1·d^-1、炒川楝子高剂量组(10 g·kg^-1·d^-1、炒川楝子中剂量组(5 g·kg^-1·d^-1、炒川楝子低剂量组(2.5 g·kg^-1·d^-1,各剂量组连续90 d灌胃,每日1次。每周测定大鼠体质量,并根据体质量调整给药量,给药期间每日观察大鼠的活动及行为,记录变化情况。灌胃结束后,检测血清和尿液肝肾功能相关的生化指标,包括丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、尿素氮(BUN)、肌酐(Cr)及N-乙酰-β-氨基葡萄糖苷酶(NAG),并观察肝肾组织的病理改变。结果 :灌胃90 d后,与对照组相比各组血清ALT、AST、Cr、BUN和尿液NAG均升高,病理检查可见不同程度的肝脏、肾脏病理组织损伤,且生化指标的升高值和病理切片的损伤情况与炒制和给药剂量呈现一定的相关性。结论 :川楝子大剂量长期给药具有一定肝肾毒性,且存在剂量依赖性,川楝子炒制后可降低肝肾毒性反应。
Objective :To observe the damage of liver and kidney of rats under long-term large dose administration of Toosendan fructus. And investigate the effects of stir-frying Toosendan fructuson liver and kidney toxicity of rats. Methods :A total of 50 male Wistar rats were divided into normal group,high does of unprocessed Toosendan fructus(10 g·kg^-1·d^-1,middle does of unprocessed Toosendan fructus(5 g·kg^-1·d^-1,low does of unprocessed Toosendan fructus(2.5 g·kg^-1·d^-1,high does of processed Toosendan fructus(10 g·kg^-1·d^-1,middle does of processed Toosendan fructus(5 g·kg^-1·d^-1,low does of processed Toosendan fructus(2.5 g·kg^-1·d^-1according to a random number table method. Every dose of group set gavage for 12 weeks,once daily,body weight was measured every week,and the dosage adjusted in real time according to the changes. And observed and record the activities and behavior in rats,daily. After the end of gavage,detectioned the serum and urine biochemical indicators which related to liver and kidney function,including alanine aminotransferase(ALT),aspartate aminotransferase(AST),blood urea nitrogen(BUN),creatinine(Cr)and N-acetyl-β-D-glucosaminidase(NAG). And pathological changes of liver and kidney were observed. Results :By gavage for 90 days,compared with the control group,serum ALT,AST,Cr,BUN and urine NAG increased in all groups,pathological examination showed different degree of liver and kidney pathology tissue damage,and the value of biochemical indicators and pathological sections has a certain correlation with the processing and administration. Conclusion :When administered with large dose and long term,Toosendan fructus has toxicity to liver and kidney,and it showed dose dependent. That toxicity can be decreased after the Toosendan fructus being processed.
