摘要
目的研究转录因子NF-E2相关因子(Nrf2)在间歇性低氧(IH)对大鼠心肌缺血再灌注(I/R)损伤的影响。方法制作成年SD大鼠心肌I/R模型,48只大鼠分为随机3组:假手术组(n=16)、I/R组(n=16)、IH+I/R组(n=16)。假手术组将生理盐水注进尾静脉,低氧处理(先给予2 min 6%~8%低氧,再进行3 min氧和处理,每次循环5次)制作IH动物模型,垫扎球囊法制作心肌I/R动物模型。术后检测心肌丙二醛(MDA)、谷胱甘肽(GSH)、肌酸激酶同工酶(CK-MB)含量。术后48 h处死,通过Western blot法检测Nrf2蛋白表达。结果 I/R组及IH+I/R组的心肌细胞MDA含量分别为16.82±6.26 nmol/L、7.53±4.18 nmol/L,GSH含量分别为132.17±3.54 mg/L、172.15±5.63 mg/L,CK-MB含量分别为2 601±3.75 U/L、1 538±2.87 U/L。I/R组及IH+I/R组Nrf2蛋白表达分别为0.217±1.326、0.872±4.538(P<0.05)。与假手术组比较,I/R组CK-MB、MDA升高,GSH含量、Nrf2蛋白表达降低,差异均具有统计学意义(P<0.05);与I/R组比较,IH+I/R组MDA、CK-MB含量降低,GSH含量、Nfr2蛋白表达升高,差异均具有统计学意义(P<0.05)。结论间歇性低氧在大鼠心肌缺血损伤中具有心脏保护作用,其保护机制与Nrf2表达升高有关。
Objective To investigate the effects of nuclear factor - erythroid related factor 2 on heart ischemic/ reperfusion myocardial damage in Intermittent Hypoxia. Methods The ischemia/ reperfusion model of adult SD rats were established,Fourty - eight rats were randomly divided into 3 groups:sham - operated group(n = 16);myocardial ischemia reperfusion group (I/ R)(n = 16),I/ R + Intermittent Hypoxia group (n = 16). The sham operation group was injected with saline into the tail vein,hypoxia treatment (given 2 min 6% ~ 8% hypoxia,followed by 3 min oxygen treatment,each cycle 5 times)to make IH animal model,Law made myocardial I/ R animal model. The Malondialdehyde(MDA), glutathione (GSH),creatine kinase - myocardial isoenzyme C(CK - MB)expression in tissues were detected,and the expressions of Nrf2 were determined by Westen blot. Results The MDA of I/ R group,I/ R + intermittent hypoxia group was 16. 82 ± 6. 26 nmol/ L,7. 53 ± 4. 18 nmol/ L respectively;the GSH of I/ R group,I/ R + intermittent hypoxia group was 132. 17 ± 3. 54 mg/ L,172. 15 ± 5. 63 mg/ L espectively. The CK - MB of I/ R group,I/ R + intermittent hypoxia group was 2601 ± 3. 75 U/ L,1538 ± 2. 87 U/ L respectively. The expression of Nrf2 in the I/ R group, I/ R + intermittent hypoxia group was 0. 217 ± 1. 326,0. 872 ± 4. 538 respectively. The levels of CK - MB,MDA,GSH and Nrf2 in I/ R group were significantly lower than those in sham operation group (P 〈 0. 05). Compared with I/ R group,IH + I/ R group the contents of MDA and CK- MB were decreased,GSH content and Nfr2 protein expression were increased (P 〈 0. 05). Conclusion Intermittent hypoxia has cardiopro-tective effects on myocardial ischemia in rats,and its protective mechanism is related to the increased expression of Nrf2.
